高效液相色谱法测定石榴皮中没食子酸的含量

摘　　要：目的测定石榴皮中没食子酸的含量。方法采用高效液相色谱法，Zorbax-C18色谱柱，流动相水-二甲基甲酰胺-冰醋酸（99．82-0．15-0．03），检测波长270nm，流速1mL·min^-1，柱温为室温。结果在0．081～0．811μg·mL^-1范围内，没食予酸峰面积（Y）与其进样量（X）呈良好的线性关系（r=0．9999），平均日内、日间RSD为1．01％和1．60％（n=6），不同浓度的平均回收率为98．35％，RSD=1．63％（n=6）。结论该法快速简便，稳定可靠，专属性强。OBJECTIVE To determine the content of gallic acid in megranate＇s-pericarp with HPLC. METHODS A zorbax C18 column （250mm×4.6mm, 5μm） was used. The mobile phase was water-dimethyformamide-glacial acetic acid （99.82 ： 0.15 ： 0.03）, and detected at 270nm; with flow rate of 1.0mL·min^-1, and the temperature of column was at room temperature. RESULTS The linearity of gallic acid was over the range of 0.081-0.811 μg·mL^-1 with the correlation coefficient 0.999 9. The average recoveries of different concentration was 98.35% with RSD 1.63% （n = 6）; the within-day and between-day RSD were 1.01% and 1.60% （n = 6）, respectivelity. CONCLUSION The method is convenient, stable, reliable and special for the dermination of gallic acid in Megranate＇s-pericarp.
