摘  要:目的建立组织芯片的手工制备技术并探讨其用于肝癌研究的可行性。方法利用改进的腰穿针手工制作组织芯片,进行免疫组织化学、组织化学染色和常规H-E染色,将组织芯片和普通切片的免疫组织化学结果对比观察。结果各种染色切片上的组织芯片基本符合观察需求,组织芯片和普通切片免疫组织化学结果基本一致。结论组织芯片具有高通量、经济节约、可比性和重复性强的特点,为高效快速地进行科研和临床工作提供一个新的技术平台。手工制备组织芯片简单易行,费用低廉,易于推广。Objective To construct a kind of tissue microarrays chip for applied in hepatocellular carcinoma researches. Methods Tissue microarrays chip were constructed manually with needles puncture in waxy matrix. Immunohistochemical, histochemical and haematoxylineosin staining were performed on the sections. The results were compared with normal sections stained by immunohistochemical. Results The cores on slides were well preserved and basically met the requirements of research. There were no significant differences between the results of tissue microarrays and normal sections that were stained by immunohistochemistry. Conclusion Tissue microarrays have characters of high-throughput, economy, comparability and representing. They provided a novel technical platform for carrying out researches and clinical routines effectively. Construction of tissue microarrays manually is an easy and effective method and is easy to apply.
