摘 要:目的:利用内皮素-1(endothelin-1,ET-1)建立一种操作简单、稳定性和重复性好的大鼠局灶性脑缺血再灌注模型并观察高压氧(hyperbaric oxygen,HBO)治疗的效果。方法:动物麻醉后,将3μl ET-1(60 pmol/μl)注射到大鼠大脑中动脉附近,建立大鼠局灶性脑缺血再灌注模型。ET-1+HBO组大鼠术后1 h给予HBO[100%O2,2.5 atmosphere absolute(ATA),2 h]治疗;ET-1组不予治疗。所有动物在术后4、8和24 h进行神经学评分,24 h断头取脑,测定左右半球脑含水量,进行TTC和H-E染色。结果:ET-1诱导的大鼠局灶性脑缺血模型成功率较高(20/24),梗死体积稳定,重复性好。经HBO治疗后,ET-1+HBO组与ET-1组相比,4、8和24 h的神经学评分均明显改善(4 h和8 h有P<0.05,24 h有P<0.01),脑含水量显著降低(P<0.01),梗死体积显著缩小(P<0.05),H-E染色示脑损伤明显减轻。结论:ET-1诱导的大鼠局灶性脑缺血模型是研究人类脑缺血疾病较理想的实验动物模型,HBO治疗能明显减轻缺血性脑损伤。Objective:To establish a focal cerebral ischemia/reperfusion (FCI/R) model with endothelin-1 (ET-1) in rats and to observe the effectiveness of hyperbaric oxygen (HBO) in treatment of this model. Methods: Three μl ET-1 (60 pmol per μl ) was injected into the site adjacent to rat middle cerebral artery (MCA) under anesthesia to induce FCI/R model. HBO (100% O2, 2.5 ATA for 2 h) was applied at 1 h after ET-1 injection in ET-1+HBO group; rats in FCI/R model group received no treatment. Neurological scores were determined at 4 h, 8 h, and 24 h after injection. Animals were all killed at 24 h to harvest brain tissues for water content assessment and TTC, H-E staining. Results: ET-1 had a high successful rate in inducing FCI/R model in rats, with stable infraction area and good repeatability. Animals in ET-1+ HBO group had better neurological outcomes than controls at all the 3 time points (P〈0.05 at 4 h and 8 h, P〈0.01 at 24 h). HBO treatment significantly decreased the brain water content (P〈0.01), reduced the infarction areas (P〈0.05), and obviously alleviated the cerebral lesions as showed by H-E staining. Conclusion: The ET-1-induced FCI/R model provides an ideal model for studying human cerebral ischemia diseases. HBO (100% O2, 2.5 ATA for 2 h) treatment administrated 1 h after focal cerebral ischemia can improve the neurological outcomes of the model animals, reduce the infarction areas, and alleviate ischemic brain lesion.
