不含二甲苯封片胶在冰冻快速制片中的应用

摘　　要：目的探讨不含二甲苯封片胶在冰冻快速制片中的应用价值及其与透明剂的正确搭配使用。方法33例临床手术切除新鲜标本各冰冻连续切片4张,分四组按冰冻快速制片程序制片。以中性树胶与二甲苯搭配为对照,观察不含二甲苯封片胶跟“TO”透明剂搭配、跟无毒透明剂搭配以及跟“TO”透明剂和无毒透明剂联合搭配的制片质量结果。制片质量结果以显微镜观察其即时效果及放置1周、放置1个月并不时暴露于阳光的保存效果。结果整个制片过程约15min。各组搭配制片均得到良好效果,组织细胞结构清晰,染色鲜艳,红蓝分明,核质对比良好。在封片整洁性及不产生刺激性气味等方面以不含二甲苯封片胶跟无毒透明剂搭配、跟“TO”透明剂和无毒透明剂联合搭配两组制片效果更佳。放置1周、放置1个月并不时暴露于阳光的保存效果良好。结论不含二甲苯封片胶在冰冻快速制片中有良好的应用价值,对改善制片和阅片环境及提高制片综合效果有重要意义,但须与透明剂正确搭配使用,才能达到应有效果。Objective To study the value of the non xylene sealant paired with clearer that used for coverslipping in frozen section. Methods 33 freshness samples came from surgical operation, the tissues from the sample were cut for four sections, that would be stained by four methods according to frozen section procedure. The control group were used neutrality rubber paired with xylene,experimental groups were used non-xylene sealant paired with＂TO＂clearer, paired with non poisonous clearer,and paired with＂TO＇clearer combination with non poisonous clearer. Observed by microscope immediately, after one week and after one month with exposured under the shine now and then. Results The frozen section procedure need fifteen minutes, histocyte structure was in focus, the nucleus and cytoplasm were clearly and constructed well. From tidiness and no irritant gas the group which usednon-xylene sealant paired with non poisonous clearer or paired with＂TO＇clearer combination with non poisonous clearer were better. After one week or one month with exposured under shine were reserved well. Conclusion Non-xylene sealant is applied cost forcoverslipping in frozen section, and is significance value in improving environment of staining and reading slides, that to be used correctly pairing with the clearer will be more fruitful.
