摘  要:目的研究大鼠实验性脑梗死周边区(A区)星形胶质细胞与突触的关系及其对运动功能恢复的意义。方法经大脑中动脉闭塞(MACO)制成脑梗死模型后,随机分90只大鼠为抑制、盐水和对照组,以横木行走试验(BWT)评定运动功能,每组在7、21、42 d应用电镜和免疫组织化学方法观察各组大鼠A区星形胶质细胞、突触、胶质纤维酸性蛋白(GFAP)和突触素(SYP)表达值的变化。结果3、6周末,抑制组大鼠运动功能、A区突触和星形胶质细胞数目及GFAP和SYP表达值均明显较盐水和对照组差(P<0.01)。结论脑梗死大鼠A区星形胶质细胞可促进突触数目增多、功能增强,这可能是其运动功能恢复的重要机制。Objective To investigate the relationship between astrocytes and synapse structure in the region around the infarction area (area A) and the roles of the astrocytes on the recovery of motor skills in the experimental rats with acute cerebral infarction, Methods Murine experimental infarction model was established by middle cerebral artery occlusion(MACO). 90 rats were divided into 3 groups randomly: inhibitory,saline and control group. On day 7, 21, and 42, the motor skills of the rats were examined via beam walking test(BWT) ; and the astrocytes ,synapse, and the expressions of glial fibrillary acidic protein (GFAP) and synaptophysin(SYP) were analyzed by immunohistochemistry staining and electron microscopy. Results At the end of 3rd and 6th week, the recovery of motor skills in the inhibitory group was significantly slower than that in the control or the saline group ( P 〈0. 01 ). The number of synapses and astrocytes in area A and the expression levels of GFAP and SYP were notably lower in the inhibitory group than those in the control or the saline group( P 〈 0. 01 ). Conclusions The increase of astrocyte number and activity is greatly associated with the number and function of synapses in area A ,which may play an important role on the recovery of motor skills in the experimental rats with acute cerebral infarction.
