摘  要:目的:比较新型光敏剂CPD4和CPD6对小鼠S180移植肉瘤的光动力学疗效。方法:给昆明小鼠接种S180肉瘤,建立移植肿瘤模型。将模型小鼠随机分成7组(n=8),分别为CPD4和CPD6中、低剂量组,血卟啉组,生理盐水对照组及未经任何治疗组。尾静脉注射给药,给药后20min以波长670nm激光垂直照射瘤区20min(血卟啉组照射60min)。治疗后30d将各组小鼠断颈处死,取瘤称质量并计算抑瘤率,做病理切片、常规染色,镜下观察形态学差异。结果:CPD4中、低剂量组的抑瘤率分别为54.5%,97.9%;CPD6低剂量组的抑瘤率为46.9%;血卟啉组的抑瘤率为55.8%,镜下观察各组切片形态学差异明显。CPD4低剂量组的疗效高于血卟啉组,亦高于同剂量CPD6组。CPD6低剂量组疗效与血卟啉组相近。结论:光动力学治疗对小鼠S180移植肉瘤有抑制作用,该研究可为生产治疗恶性肿瘤的新药提供科学的实验依据。Objective: To compare the photodynamic effects (pDrF) of new photosensitizers CPD4 and CPD6 on sarcoma S180 transplanted in mice. Methods: The animal model of tumor was developed by inoculating Kunming mice with sarcoma S180 The tumor-transplanted mice were randomly divided into seven groups and eight mice each one including the low and medium dose group of CPD4 and CPD6, the hematoporphyrin dirivative (HpD) group, the normal saline group and the control group. The photosensitizers were given by means of vena caudalis then tumors were irradiated vertically for twenty minutes with laser whose wave length was 670 nm after twenty minutes administration (sixty minutes administration for the hematoporphyrin dirivative group). Thirty days after the photodynamic therapy, all of the mice were killed and the tumours were extirpated. The tumor inhibitory rate was calculated ,the tumor samples were sliced up,HE dye and made a morphological difference research under the microscope. Results: The tumor inhibitory rates (TIR) of new photosensitizer CPD4 were 54.5% ,97.9% at the medium and low dose group respectively; the TIR of CPD6 was 46.9% at the low dose group. The TIR of the HpD was 55.8%. The morphological differences of the slices were evident in different groups.The low dosage of CPD4 had a better theraputic effect than HpD and CPD6 at the same dose. The photodynamic effect of CPD6 at low dose is similar to that of HpD. Conclusion: The photodynamic effect can inhibit the growth of sarcoma S180 transplanted in mice. This result may provide scientific experimental evidence for working out new agents against malignant tumor.
