摘 要:以对硝基苯磷酸二钠为底物,研究了大豆体外酸性磷酸酯酶的酶促反应动力学和氟化钠对该酶的抑制动力学特性,实验结果表明,大豆酸性磷酸酯酶的酶促反应初速度时间范围为0~20min,最适pH5.6,最适反应温度为37℃,米氏常数(Km)为2.78×10-4mol/L,最适反应速度(Vm)为1.15×10-8mol/min.NaF对大豆体外酸性磷酸酯酶有明显抑制作用,属于非竞争性可逆抑制类型,其抑制常数(Ki)为1.53×10-3mol/L.此外,还探讨了NaF的抑制机理.In this experiment we studied the kinetics of enzyme catalyzed reaction of acid phosphatase extracted from soybean and the inhibition characteristic of the reaction kinetics affected by sodium fluoride.The experimental results showed that for the enzymatic reaction of acid phosphatase of soybean,the initial rate lasted for 20 min,the most optimum conditions were pH 5.6 and 37 ℃,Michaelis Menton constant (K m) was 2.78×10 -4 mol/L,the maximum reaction velocity (V m) was 1.15×10 -8 mol/min. The results also demonstrated clearly that,NaF had great inhibition effect on acid phosphatase activity,this inhibition effect belonged to non competiton type,and the inhibition constant (K i) was 1.53×10 -3 mol/L.The inhibition mechanism of sodium fluoride was also discussed in this paper.
