摘 要:本研究主要包括:(1)建立了猪MHS基因检测的PCR-RFLP方法。该方法具有准确、快速和无侵害的特点;(2)利用猪MHS基因的PCR-RFLP方法对中国现有猪种20个群体的MHS基因座位的基因和基因型频率进行了检测。结果表明,属于中国的本地品种五指山猪、香猪,引进品种约克夏猪,均不含有MHS基因;在中国本地品种二花脸猪和民猪中,发现存在MHS基因,且民猪的频率较高(0.1562);培育品种北京花猪,引进品种丹系长白猪和杜洛克猪,均有MHS基因;引进品种皮特兰猪、比系长白猪,MHS基因频率最高,分别为0.9520和0.9432;(3)猪RYR1/CRC基因部分DNA序列品种间比较,MHSNN香猪和二花脸猪的1843位为“C”,而MHSnn为“T”;This study consists of three parts: (1) A PCR RFLP test was established to identify the gene at MHS locus. The DNA base test of MHS gene is a rapid, accurate and uninvasive method. (2) The gene and genotype frequencies of MHS locus in 20 pig populations in China were investigated by the PCR RFLP test. MHS gene exists in Erhualian and Min Pig of China native breed; Beijing Hua Pig of China developed breed; Pietrain, Duroc, Belgian Landrace and Danish Landrace of China exotic breed; Line A, B and C of Dekalb Pig, being not found in Wuzisan,Xiangzhu,Yorkshire,Beijing Heizhu and Dekalb E,F lines. In MHS gene frequency, the highest populations are Pietrain (0.951 9) and Belgian Landrace (0.943 2); the middle ones are Min Pig (0.156 2), Line B (0.171 6) and C (0.084 6) of Dekalb; the lower ones are Duroc (0.04), Danish Landrace (0.035 7) and Beijing Hua Pig (0.036 5); and the lowest is Erhualian Pig (0.005 7). (3) Comparison among the DNA sequences of different breeds. The T base is at the 1843 site of MHS gene of MHS nn Pietrain Pig, but C in the same site of MHS NN Xiang Pig and Erhualian Pig. In 1 795 ̄1 928 coding region, the some other base variations are also found.
