摘  要:目的研究丹参对动脉粥样硬化的预防作用机制。方法建立家兔动脉粥样硬化模型,比较正常组、AS组及丹参组动脉粥样硬化斑块形成的变化;检测各组血清脂质水平;免疫组化结合RT-PCR法分析Bcl-2、Bax及MCP-1在粥样硬化斑块中的表达。结果丹参组主动脉粥样斑块面积和血清TG水平低于AS组(P<0.01);免疫组化显示,AS组Bcl-2、Bax及MCP-1阳性细胞表达与正常组有显著性差异。而丹参的干预,可以调节Bax/Bcl-2阳性细胞表达率,同时下调MCP-1的蛋白表达。结论丹参可抑制动脉粥样硬化斑块的形成,调节Bcl-2/Bax表达及炎症机制参与可能是其抗动脉粥样硬化的机制之一。Objective It is to explore the mechanism of preventing function of Danshen on atherosclerosis (AS). Methods AS models of rabbit were made, and the changes of AS plaques in normal group, AS group and Danshen group were compared. The level of serum fat in every group was determined, and the expression of Bcl - 2, Bax and MCP - 1 in AS plaques was analyzed with immunohistochemistry method and RT - PCR method. Results The areas of AS plaques and the content of serum TG were less in Danshen group than those in AS group (P 〈 0.01 ). Analysis of immunohistochemistry showed that there was a significant difference in the expression of positive cells of Bcl - 2, Bax and MCP - 1 between AS group and normal group. Bax/Bcl - 2 was decreased and MCP - 1 protein was inhibited in Danshen group. Conclusion Danshen can inhibit the formation of AS plaques. Improving the expression of Bcl - 2/Bax and inflammation function may be one of the mechanisms.
