摘　　要：目的观察m ompS基因DNA疫苗对嗜肺军团菌感染的免疫保护作用。方法将18只BALB/c雌性小鼠随机分为3组,pcDNA 3.1-m ompS实验组股四头肌肌肉接种pcDNA 3.1-m om opS质粒100μg,两周后加强免疫1次,2个对照组分别肌肉注射等体积的生理盐水和空质粒,加强免疫后3周小鼠鼻腔滴注嗜肺军团菌L 1型菌液进行攻击感染,感染4周后,检查小鼠肺组织带菌量,观察肺组织病理形态学改变。结果pcDNA 3.1-m ompS免疫组肺组织带菌量少于生理盐水对照组和空质粒对照组(P<0.01);与生理盐水对照组和空质粒对照组比较,pcDNA 3.1-m ompS免疫组肺组织病理改变轻微。结论由m ompS基因构建的嗜肺军团菌DNA疫苗能诱导小鼠产生保护性免疫。Objective To observe the immunoprotection effect of DNA vaccine containing mompS gene against legionella pneumophila infection. Methods Eighteen female BABL/c mice of 6-8 weeks old were divided randomly into three groups, in which the pcDNA3. 1-mompS group was immunized intramuscularly with recombinant plasmid pcDNA3.1-mompS; the blank plasmid group and normal saline group were injected with blank plasmid pcDNA3.1 （＋） and normal saline respectively. Immunization was boosted twice with the same dosages at two weeks interval. Three weeks after last immunization, all the mice were challenged with legionella pneumophila serotype 1. Four weeks after challenging, the bacterial colony counts and the pathological changes in lungs were determined and observed to evaluate the protective immunity of DNA vaccine. Results The bacterial colony of legionella pneumophila in the pcDNA3. 1- mompS groupwas significantly lower than that of the normal saline group and the pcDNA3.1 （＋） group （P〈0. 05）. The pathological changes of lungs in the pcDNA3.1-rnornpS group were mild compared with that of the blank pcNDA3. 1 （＋） group and normal saline group. Conclusion Protective immunity against legionela pneumophila can be induced by the DNA vaccine containing mompS gene, which may provide a good experimental basis for further developing DNA vaccine against legionella pneurnophila.
