摘 要:目的探讨环孢霉素A滴眼液对结膜干燥症老鼠杯状细胞与结膜上皮粘蛋白5AC(MUC5AC)表达的影响。方法30只C57BL6小鼠皮下注射东莨菪碱12d制造结膜干燥症模型鼠,十只为干眼对照组不使用局部治疗,十只小鼠使用1μL0.05%环孢霉素A(CsA)滴眼每日3次(干眼+CsA组),十只小鼠使用1μL蓖麻油滴眼每日3次(干眼+载体组)。另取十只为正常组,作为未作处理的对照。采用PAS染色、免疫组织化学及Westernblot方法,对上述4组小鼠结膜上皮细胞中的杯状细胞与MUC5AC的表达进行检测。结果杯状细胞数目、MUC5AC免疫组化及Westernblot表达在正常组和干眼+CsA组均高于眼对照组和干眼+载体组(P均<0.01);各组杯状细胞数目与MUC5AC表达均呈正相关(P均<0.01)。结论CsA可稳定膜结膜杯状细胞,促进黏蛋白分泌,从而维持泪腺分泌的基本功能。[Objectivel To study the effect of the number of globet cells and mucin expression in experimental murine xerophthalamia conjunctlval epithelium with topical cyclosporine (CsA) treatment. [Methods] Dry eye was induced in 3 groups of C57BL6 mice by subcutaneous injection of scopolamine TID and exposure to an air draft and low-humidity environment for 16 hours per day for 12 days. The dry cyc control group received no topical treatment; a second group received 1 μL of 0.05% CsA topically TID (dry eye + CsA); and the third group received 1 μL of the castor oil vehicle of CsA topically TID (dry eye + vehicle). Normal mice were used as untreated controls. 12 days later, the mice were killed, and their conjunctivas were excised. Their conjunctival goblet cell number was counted in tissue sections stained with period acid schiff (PAS) reagent. Their conjunctiva epithelium had been investigated by immunohistochcmical staining to detect the goblet cells, and the expression of Muc SAC had been investigated by immunohistochemical and Western blotting methods. [Results] Compared with dry eye control and dry eye mice + vehicle groups, the number of conjunctival epithelial goblet cells and the expressions of MUC SAC was significantly greater in the untreated controls and dry cyc mice receiving CsA groups (P 〈 0.01 for both groups). There was no significant difference in the number of conjunctival epithelial goblet cells between the dry eye control and dry eye + vehicle group. So was the number of conjunctival epithelial goblet cells between the normal and the dry eye + CsA group. There was a significant positive correlation between goblet cell numbers and the cell numbers that expressed MUC 5AC. (P 〈 0.01 for both groups). [Conclusion] Topical CsA significantly stable conjunctival epithelial globet cells, promote the secretion of MUC SAC, and keep the secretion function of lacrimal gland.
