摘 要:目的:探讨不同浓度的胰岛素和人参皂甙Rg1对体外3T3-L1脂肪细胞脂联素(ad iponectin)mRNA表达的影响。方法:通过不同浓度胰岛素和Rg1与3T3-L1脂肪细胞共同培养,以β-actin为内对照,半定量逆转录PCR法测定脂联素mRNA表达。结果:随着胰岛素浓度的升高,脂联素mRNA表达逐渐降低,在胰岛素浓度≥100 nmol/L时具有显著差异(P<0.05);40 mg/L Rg1能有效逆转高胰岛素对脂联素mRNA表达的抑制作用(P<0.05)。结论:体外高胰岛素水平可使3T3-L1细胞脂联素表达下降,Rg1可逆转体外高胰岛素降低脂联素表达的作用。AIM: To explore the effect of different concentrations of insulin and ginsenoside Rgl on the expression of adiponectin mRNA in 3T3 - L1 adipocytes. METHODS: After 3T3 - L1 adipocytes were incubated with different concentrations of insulin and ginsenoside Rgl for 48 hours, the level of adiponectin mRNA in 3T3 -L1 adipocytes was determined by reverse transcription - PCR taking 13 - actin as internal reference. RESULTS : With the increase in insulin concentration, the expression of adiponectin mRNA in 3T3 - L1 adipocytes was decreased ( P 〈 0. 05 ), and 40 mg/L Rgl reversed the decrease in adiponectin mRNA expression (P 〈0. 05). CONCLUSION: In vitro, insulin at high concentration decreases the expression of adiponectin mRNA in 3T3 -L1 adipocytes, which is reversed by treatment with ginsenoside Rgl.
