摘  要:目的比较分析灭活贝氏柯克斯体全细胞(WC)和氯仿-甲醇提取WC的残存组分(CMR)Q热疫苗的脂肪酸种类及含量差异。方法制备WC和CMR,采用高压气相色谱检测WC和CMR脂肪酸种类及相对含量。结果WC经氯仿-甲醇回馏处理后得到的不溶性组分CMR。气相色谱分析发现CMR较WC减少了11种脂肪酸,其中4种为不饱和脂肪酸,4种为支链脂肪酸,70%为C原子数≥18的较长链的脂肪酸。另外,两种疫苗的支链脂肪酸与直链脂肪酸、不饱和脂肪酸与饱和脂肪酸的比值也明显不同。结论灭活贝氏柯克斯体疫苗经氯仿-甲醇提取后,除去了它的多种脂肪酸或减少某些脂肪酸含量,这些脂肪酸的消除与减少可能是CMR疫苗诱导的局部和全身不良反应消失或减轻的主要原因。To investigate the differences in fatty acid contents within the whole cells (WC) and the residues extracted with chloroform and methanol (CMR), the compositions and contents of fatty acids were analyzed by gas chromatography. It was found that there was a reduction of 11 fatty acids in CMR in comparison with those of WC as demonstrated by gas chromatography analysis, in which four of these 11 fatty acids appeared to be unsaturated fatty acids; the other four were branchedchain fatty acids; and 70% of them were long-chain fatty acids with 18-C-atoms or more. In addition, the ratios of the branched-chain fatty acids to linear chaln-fatty acids and that of the unsaturated fatty acids to saturated fatty acids were also significantly different between CMR and WC. These results suggest that many fatty acids vanish or reduce in quantity after extraction with chloroform and methanol from the whole cells, which may be the major cause to explain the reduction of the adverse reactions induced by immunization with CMR vaccine against coxiellosis.
