摘 要:目的:应用高效液相色谱分析技术对次声作用后小鼠睾丸细胞基因组DNACpG甲基化的变化进行分析,以研究次声作用与表遗传学改变的关系.方法:采用8Hz,130dB的次声分别作用于1,7和14d组3个实验组,各雄性Balb/c小鼠20只,每日次声暴露2h.同时设对照组,并利用高效液相色谱分析技术对实验组与对照组的小鼠睾丸细胞基因组DNACpG甲基化水平进行检测.结果:次声暴露1,7和14d组的小鼠睾丸细胞基因组DNACpG甲基化程度均低于对照组,并随次声作用时间的变化而改变,分别为0.0150±0.0127vs0.0235±0.0062,0.0140±0.0030vs0.0273±0.0040,0.0150±0.0096vs0.0250±0.0049(P=0.000).结论:次声作用可致睾丸细胞基因组DNACpG甲基化的改变,与次声暴露时间有关.AIM: To investigate the changes of genome DNA methylation of testicular cells in mice with high performance liquid chromatography (HPLC) after infrasound exposure, and to explore the relations between infrasound and epigenetics. METHODS: The BALB/c mice were exposed to infrasound (8 Hz/130 dB) for 1,7 and 14 d, respectively, 2 h daily, and the control groups were unexposed. Then the changes of genome DNA methylation of testicular cells in the mice were analyzed with HPLC. RESULTS : The levels of the genome DNA methylation of testicular cell in exposed groups were all significantly lower than those in control groups as time went on (1 d, 0.0150±0.0127 vs 0.0235±0. 0062; 7 d, 0. 0140±0. 0030 vs 0. 0273 ±0.0040; 14d, 0.0150 ±0.0096 vs 0.0250 ±0.0049. P =0.000). CONCLUSION: The infrasound markedly affects the genome DNA methylation of testicular cells. These changes are positively associated with the infrasound exposure time.
