摘 要:比较了不同生长调节剂及其种类浓度和培养时间对‘富有’和‘次郎’两个甜柿(DiospyroskakiThunb.)品种的叶片愈伤组织诱导和不定芽再生的影响。TDZ诱导叶片愈伤组织和不定芽再生的能力很强,在含ZT0·5mg·L-1的培养基中,愈伤组织的形成率达96%,愈伤组织不定芽分化率达80%以上,由0·5mg·L-1TDZ诱导形成的富有和次郎愈伤组织,在含有ZT0·5mg·L-1的1/2DKW培养基培养30d,不定芽分化率均达96%以上,平均芽数分别为7·8和5·4,所得再生苗的生根率近75%。Studies were carried out on high-frequency shoot regeneration from leaves of two Japanese non-astringent persimmon. The effects of growth regulator and cultural time on Fuyu and Jiro, the adventitious shoot regeneration from leaves were investigated. The results showed that TDZ was a strong and efficient hormone inducing adventitious shoot and calli formation. The best regeneration was achieved in 1/2DKW + TDZ 0. 5 mg · L^-1 + ZT 1.0 mg · L^-1, in which callus percentage was 96% and shooting was near above 80%. After 30 day culture, the calli with small shooting were transferred to 1/2DKW medium supplemented with 0. 5 mg · L^-1 zeatin and 0. 1 mg · L^-1 IAA. Shooting regeneration rate were more than 96%, and average adventitious shoots per explant were 7.8 and 5.4 respectively for Fuyu and Jiro after 30 days culture. Rooting percentage from regenerated shoots was about 75%.
