摘 要:目的建立稳定的大鼠供肝冷保存肝移植模型,为冷保存再灌注损伤及再生研究奠定基础。方法大鼠原位肝移植模型参照Kamada的双袖套法并加以改进。雄性SD大鼠分为4组,观察不同的冷保存时间对大鼠生存率及损伤与再生状况的影响。结果模型的平均无肝期为15min左右,肝移植对照组、冷冻保存8、12、16h组术后168h存活率分别为100%、80%、50%、20%。冷冻保存12h组与肝移植对照组相比,大鼠存活率明显降低(P<0.05)。冷冻保存12h组肝脏组织术后24h可观察到明显的片状坏死,继而肝细胞有丝分裂相显著增多。结论对SD大鼠而言,冷保存12h的供肝肝移植是研究肝脏损伤与再生修复的良好模型。Objective To study the impact of different cryopreservation time on the injury and regeneration of the donor liver after transplantation. Methods Orthotopic liver transplantation was performed using the two-cuff technique described by Kamada with modification. According to the different cryopreservation time of the donor liver in the University of Wisconsin (UW) solution, male SD rat recipients were randomly divided into four groups: group 1 (1 h), group 2 (8 h), group 3 (12 h) and group4 (16 h), with 10 recipients and 10 donors in each group. After transplantation, the operation parameters, survival rate, liver injury and regeneration of the recipients were observed. Results The average anhepatic time was consistently kept below 15 min. The 168-hour survival rate was 100% , 80% , 50% and 20% in group 1,2, 3 and 4, respectively. The survival rate in group 3 was significantly lower than that of group 1 ( P 〈 0.05 ). Submassive confluent necrosis and increased mitotic figures could be observed in group 3. Conclusion The SD rat that received the liver subjected to 12-hour cryopreservation may be the ideal model for the research of liver injury and regeneration during the early period after liver transplantation.
