摘 要:目的:初步探讨刚地弓形虫低毒(T.gondii)DX株-速殖体可溶性抗原(STAg)对小胶质细胞的影响。方法:采用流式细胞术,对不同处理组小鼠小胶质细胞及神经元细胞的凋亡情况进行了分析。并应用原位TUNEL法检测了不同处理组小鼠的小胶质细胞凋亡情况。结果:Tp、TpTi(颅内注入T.gondii)鼠小胶质细胞凋亡数均值约为4个/hpf,高于正常鼠。Tp鼠,凋亡神经元占66%。Tp、TpTi鼠小胶质细胞无NO阻断剂(1-NAME)处理:神经元凋亡分别占60%和71%;而Tp、TpTi小胶质细胞有NO阻断剂(1-NAME)处理:神经元凋亡分别占20%和23%。结论:T.gondii低毒DX-STAg刺激后,所产生的小胶质细胞具有神经毒性。且NO抑制剂能显著性阻断Tp、TpTi鼠小胶质细胞对神经元的凋亡效果。Objective. To explore the influence of T. gondii (DX strain) soluble tachyzoite antigen (STAg) on mice microglia. Methods: Flow cytometry was used to detect functions of neurons from NML,Tp and TpTi mice. TUNEL was used to detect functions of microglia. Results: The numbers of apoptotic microglia in cortex in Tp and TpTi mice were about 4/hpf detected by TUNEL assay in vivo. The percen of apoptotic neurons in Tp mice was 66%. The percentage of apoptotic neurons co-cultured with Tp and TpTi microglia (without 1-NAME-treatment) were 60% and 71%, respectively. But the percent of apoptotic neurons co-cultured with Tp and TpTi microglia plus NO inhibitor (with 1-NAME-treatment) were 20% and 23%, respectively. Conclusion. T. gondii- STAg elicits neurotoxic microglia. NO inhibitor N^ω-nitro-l-arginine methyl ester (1-NAME) significantly inhibits apoptotic effect of Tp and TpTi microglia on neurons in vitro.
