摘  要:目的 用组织浴槽血管环技术研究15-酮基二十碳四烯酸(15-KETE)收缩大鼠离体肺动脉的作用及其离子通道机制。方法 16只健康的Wistar大鼠,体重为(220±20)g,随机分为两组(n=8);正常组置于正常环境中饲养(FiO2=21%),缺氧组置于缺氧的培养箱中饲养(FiO2=10%),连续饲养9d后处死,游离直径为0.5~1、0mm肺内肺动脉(PA)剪成3mm长的血管环,观察钾离子通道阻断剂、L-型钙离子通道阻断剂和无钙离子Krebs液对15-KETE诱导的正常和缺氧大鼠肺动脉环收缩的影响。结果①15-KETE以浓度(10-~10^-6mol·L^-1)依赖的方式收缩大鼠离体肺动脉环;②2mmol·L^-14-氨基吡啶(4-aminopyridione,4-AP)可明显降低15.KETE收缩大鼠离体肺动脉环的作用,正常组和缺氧组的结果相似;③10mmol·L^-1四乙胺(tetraeth—ylammonium,TEA)、10~mol·L^-1格列本脲(glyburide,GLYB)对15-KETE收缩大鼠离体肺动脉环的作用无明显影响;④10~mol·L^-1硝苯地平(nifedipine)和无钙Krebs液对10~mol·L^-1 15-KETE引起的离体肺动脉环收缩具有显著抑制作用。结论 15-KETE收缩离体肺动脉环的作用依赖于KV通道、细胞外液钙离子和L-型钙离子通道。Aim To investigate the effect of 15-ketoeicosatetraenoic acid (15-KETE) and its mechanism through ion channels on rat isolated pulmonary arterial rings by using organ bath technique. Methods Sixteen healthy Wistar rats weighing 220 ± 20 g were divided into two groups ( n = 8) : normoxia group breathing fresh air ( FiO2 = 21% ) and hypoxia group breathing hypoxic air ( FiO2 = 10% ) in a hypoxic box. Pulmonary arteries (PA) were extracted after 9 d and cut into rings (0. 5 - 1.0 mm in diameter and 3 mm in length) for organ bath experiments. Results (1) With increasing concentration from 0 to10^-6 mol · L^-1, 15-KETE increased PA rings tension gradually in a dose-dependent fashion; (2) 4-aminopyridine (2 mmol · L^-1), a Kv channel blocker significantly decreased constriction of rat isolated PA rings induced by15-KETE, and results were similar in two groups; (3) The KATP ehannel bloeker glyburide (10^-6 6mol · L^-1) and the BKca channel blocker tetraethylammonium( 10 mmol · L^-1 ) did not affect constriction of rat isolated PA rings indueed by 15-KETE; (4) The BK- Ca ehannel bloeker nifedipine ( 10^-6 mol · L^-1 ) and Ca^2+-free Krebs solution significantly decreased constrietion of rat isolated PA rings indueed by 15-KETE. Conclusion Kv ehannels play a role in eonstrietion of PA induced by 15-KETE;L-type Ca^2+ channel blocker and extraeellular ealeium ion also influenee eonstrietion of isolated PA rings induced by 15-KETE.
