摘  要:目的:探讨丹参对自发性高血压大鼠(SHR)左室肥厚及心肌细胞凋亡的作用。方法:18只引种繁殖的雄性SHR随机分成3组,每组6只:①基础组:育至第8周处死;②丹参组:从第8周给药,丹参粉针剂1g/kg.d-1,腹腔注射,第18周处死;③高血压组:用相同容量蒸馏水腹腔注射,余同丹参组。测量大鼠尾动脉收缩压(SBP)及左心室重量指数(LVM I),应用HE、VG染色,结合计算机图像分析技术,检测心肌细胞的直径(TDM)和面积(CA)、心肌组织胶原体积比例(CVF)、血管周围胶原面积与管腔面积比例(PVCA),心肌细胞凋亡的检测采用TdT介导的原位末端缺口标记法(TUNEL),Bc l-2基因的蛋白表达检测采用S-P免疫组化法。3组间各指标进行显著性检验。结果:与基础组比较,高血压组的SBP、LVM、ITDM、CA、CVF、PVCA、A l显著增加,PEI降低(P<0.05);丹参组除SBP外,各指标与基础组无差异。结论:SHR心肌细胞凋亡与左室肥厚有密切关系;长期应用丹参治疗,能显著上调SHR心肌细胞Bc l-2基因蛋白表达,抑制心肌细胞凋亡,预防高血压左室肥厚的形成。Objective : To observe the effect of Salvia mihiorrbiza on left ventricular hypertrophy(LVH) and cardiomyocyte apoptosis in spontaneously hypertensive rats(SHR). Methods: 18 male SHR were divided in 3 group sat random( n = 6) :①the base group,which were killed in the 8th week;②the Salvia group,which Salviamihiorrhiza( 1 g/kg · d^-1)were injected to intraperitoneally from the 8th week to the 18th week;the control group, which the same volume of vehicle were injected in the same manner. Systolic blood pressure(SBP) and leftventricular mass index(LVMI) were measured. Combined with computed morphometry,HE and VG were employed to evaluate the cardiomyocyte size, diameter, the collagen volume fraction ( CVF), perivascular circumferential area (PVCA). The TUNEL method was employed to evaluate cardlomyocyte apoptosis, and the immunohistochemistry method was employed to detect the protein expression of Bcl -2. Results:Compared with the base group, the SBP, LVMI, cardiomyocyte size, diameter,CVF, PCVA, and apoptosis index increased markedly in control group( P 〈 0.05). Simultaneously ,the positive expression index of Bcl-2 protein decreased significantly in control group ( P 〈0.05). Treated with Salvia mihiorrhiza,the LVH was prevented , thecardiomyocyte apoptosis decreased and the positive expression index of Bcl - 2 protein increased whereas SBP changed slightly. Conclusion:The study show that eardiomyocyte apoptosis is closely associated with LVH in SHR. Chronic use of Salvia miltiorrhiza can upgrade the protein expression of Bcl -2 gene, decrease the cardionlyocyte apoptosis and inhibit the development of LVH in spontaneously hypertensive rats.
