连钱草、人参、黄芪等组分的艾克清在小鼠体内抗肿瘤作用

摘　　要：目的:观察由连钱草、人参、黄芪、白术、茯苓、莪术等组成的方剂艾克清在小鼠体内的抗肿瘤作用。方法:实验于2005-06/2005-12在河南中医学院药理毒理学研究室完成。瘤株:小鼠肺癌(Lewis)、小鼠肉瘤(S180)、小鼠肝癌(HepA)、小鼠艾氏癌(Esc),由中国医学科学院实验动物中心引入。艾克清胶囊内容物(由河南中医学院制剂研室提供,批号20041218),配制成含0.5%的羧甲基纤维素纳混悬液0.1g/mL。取C57BL/6雌性小鼠200只,各取50只分别用于小鼠Lewis肺癌、小鼠S180肉瘤、小鼠HepA肝癌、小鼠Esc艾氏癌4种不同癌细胞瘤株的注射。将抽取的小鼠Lewis肺癌、小鼠S180肉瘤、小鼠HepA肝癌、小鼠Esc艾氏癌液体,用生理盐水1∶3稀释成1×107个细胞。在C57BL/6小鼠右腋下分别注入小鼠Lewis肺癌、小鼠S180肉瘤、小鼠HepA肝癌、小鼠Esc艾氏癌细胞悬液0.2mL。均于接种后次日,按随机抽签法分为5组,每组10只。然后给药:①艾克清小剂量组:灌胃艾克清混悬液1.0g/(kg·d)(相当于临床用量8.7倍)。②艾克清中剂量组:灌胃艾克清混悬液2.0g/(kg·d)(相当于临床用量17.4倍)。③艾克清大剂量组:灌胃艾克清混悬液4.0g/(kg·d)(相当于临床用量34.8倍)。④阳性对照组:肌肉注射环磷酰胺20mg/(kg·d)。⑤模型组:灌胃等体积的生理盐水(艾克清给药量均为等体积)。各组连续给药10d,末次给药后1h,每只C57BL/6小鼠腋下肿瘤生长明显,包膜完整,然后处死小鼠,剥离摘取每只小鼠的肿瘤称重,计算抑瘤率。抑瘤率=(对照组-给药组)/对照组×100%。结果:200只小鼠均进入结果分析。艾克清大、中、小剂量组的小鼠Lewis肺癌瘤质量显著低于模型组[(0.74±0.12),(0.89±0.13),(0.94±0.11),(1.32±0.13)g,P<0.01],抑瘤率分别为43.98%、32.32%、29.22%。艾克清大、中剂量组连续用药10d后,小鼠S180肉瘤质量显著低于模型组[(0.87±0.13),(1.06±0.22),(1.55±0.19)g,P<0.05,P<0.01],抑瘤�AIM： To observe the in vivo anti-tumor effect of Ai Keqing, a kind of Chinese herbs containing gleehomae, ginseng, radix astragali, rhizoma, poria, rhizoma zedoariae and so on in mice. METHODS： The experiment was conducted in the Department of Pharmacology and Toxicology, Henan University of Traditional Chinese Medicine from June to December 2005. The mouse lung tumor （Lewis）, mouse sarcoma （S180）, mouse liver cancer （HepA）, and mouse Ehrlich carcinoma （Esc） cell lines were supplied by the Experimental Animal Center of the Chinese Academy of Medical Sciences. The suspension of Ai Keqing capsule contents （produced by the laboratory of Chinese medicine preparation, Henan University of Traditional Chinese Medicine, batch number： 20041218） was prepared with 0.5% sodium carboxymethyl cellulose （CMC- Na） with final concentration of 0.1 g/mL. Fifty mice of 200 C57BL/6 female mice were taken, respectively for injection of four different tumor cell lines： mouse Lewis lung cancer, mouse S180 sarcoma, nlonse HepA liver cancer and mouse Esc carcinoma. Mouse Lewis lung cancer, mouse S180 sarcoma, mouse HepA liver cancer and mouse Esc carcinoma cell lines were collected and diluted at 1：3 with saline to 1×10^7 cclls/mL, The mice were injected under the fight armpit 0.2 mL suspension of four different tumor cell lines. The day after inoculation, 50 mice were administrated after randomly divided into 5 groups with 10 animals in each group： （1）Ai Keqing lowdose group： Ai Keqing suspension 1.0 g/kg per day （equivalent dose of 8.7 times of clinical dose） was infused gastrically; （2）Ai Keqing medium-dose group： Ai Keqing suspension 2.0 g/kg per day （equivalent to 17.4 times of the clinical dose）; （3）Ai Keqing high-dose group： Ai Keqing suspension 4.0 g/kg per day （equivalent dose of 34.8 times of clinical dose）; （4）positive control group： Cyclophosphamide 20 mg/kg per day was intramuscularly injected; （5）model group： Matching saline was infused, Continu
