摘 要:目的:通过体外实验初步探讨从兰科植物石斛的茎中提取的单体化合物对人肝癌细胞株FHCC-98的细胞增殖抑制作用。方法:实验于2006-04/07在解放军第四军医大学细胞工程研究中心完成。采用噻唑蓝法筛选由金钗石斛提取分离的31种单体化合物,测定其在体外对人肝癌细胞株FHCC-98的生长抑制作用,以人正常肝细胞QSG7701为阴性对照。首先利用单浓度(100μmol/L)、单时间点(48h)对化合物进行初筛,其中有3个化合物符合要求,可以进行复筛,分别为玫瑰石斛素(Crepidatin)、鼓槌联苄(Chrysotobibenzyl)、4,4’-二羟基-3,3’,5-三甲氧基二苄(Moscatilin)。复筛采用多浓度点(100,50,25,12.5,6.25μmol/L)、单时间点(48h)对化合物检测细胞毒性(IC50)。IC50值>100表示无细胞毒性。结果:玫瑰石斛素、鼓槌联苄、4,4’-二羟基-3,3’,5-三甲氧基二苄3种化合物对人肝癌细胞株FHCC-98显示不同的增殖抑制作用(IC50值分别为74.30±0.98,56.60±0.92,8.68±0.95),其中化合物4,4’-二羟基-3,3’,5-三甲氧基二苄的作用尤为明显,而3种化合物对正常细胞QSG7701基本没有毒性(IC50值>100)。结论:从金钗石斛中提取的3种单体化合物对人肝癌高侵袭细胞株的增殖具有一定抑制作用。AIM: To investigate the anti-proliferative effect of compounds isolated from Dendrobium Nobile in vitro on human hepatoma cell line FHCC-98. METHODS: The experiment was performed from April to July 2006 in Cell Engineering Research Center of Fourth Military Medical University of Chinese PLA. Compounds isolated from Dendrobium Nobile affected with inhibitory effect on growth of FHCC-98 cell line were screened by using MTr assay. The negative control was QSG7701 liver cell line. The first screening was using mono-concentration (100μmol/L), mono-point of time (48 hours). Three compounds those were Crepidatin, Chrysotobibenzyl and Moscatilin were selected from 31 compounds and re-screening was performed by multi-concentration ( 100,50,25,12.5,6.25 μmol/L), monopoint of time (48 hours), the cytotoxicity (IC50) of compounds was detected. IC50 values over 100 showed little cytotoxocity. RESULTS: Growth inhibition of crepidatin, chrysotobibenzyl and moscatilin on hepatoma cell line FHCC-98 were different (IC50 values were 74.30±0.98,56.60±0.92 and 8.68±0.95, respectively), especially Moscatilin. These three compounds showed little cytotoxicity on QSG-7701 (IC50 values over 100). CONCLUSION: Three compounds of bibenzyls from Dendrobium Nobile show growth inhibition on high invasive human hepatoma cell line.
