摘 要:目的:研究成年大鼠脑梗死后室周带神经干细胞的增殖及迁移。方法:制作大鼠脑梗死模型,将其分为脑梗死后3、5、7和14 d组,对照组为假手术组。采用免疫组化法动态检测大鼠脑内5-溴脱氧尿苷嘧啶(B rdU)、双皮质素(DCX)的表达。结果:与对照组相比,室周带B rdU阳性细胞在脑梗死后逐渐增加,并于7 d达高峰,14 d后开始下降,但仍高于正常水平。梗死后不同时间组室周带B rdU阳性细胞数均高于对照组(P<0.01)。DCX阳性细胞具有迁移细胞的形态学特征,并向损伤区迁移,于术后5 d迁移细胞的数量达高峰。结论:脑梗死可激活室周带神经干细胞原位增殖,并向损伤区迁移。Objective:To investigate the generation and migration of inherent neural stem cells in the adult rats with cerebral infarction. Methods: Rat cerebral infarction model was made and the dynamic expressions of bromodeoxyuridine (BrdU) and doublecortin (DCX) were determined by immunofluorescence staining. Results:Compared with the controls, the number of BrdU immunoreactive cells in the subventricular zone (SVZ) increased significantly (P 〈 0.01 ) after focal cerebral infarction. The expression of BrdU cells was peaked at day 7, it decreased at day 14 but remained at a level higher than that of the controls. DCX immunoreactive cells in the SVZ were morphologically migrating, they migrated to the ischemic lesion and the number was peaked at day 5 (P 〈 0.01 ). Conclusion : The results indicated that generation of inherent neural stem cells was in the SVZ and the neural stem cells migrated to the ischemic lesion area after ischemic stroke.
