摘 要:目的研究选择性COX-2抑制剂NS-398对宫颈癌细胞增殖和凋亡的影响。方法不同浓度NS-398处理体外培养的人宫颈癌细胞系Hela细胞后,采用噻唑蓝(MTT)法检测处理48h、72h后hela细胞的增殖活性,流式细胞仪测定细胞周期和细胞凋亡。结果50、100、200umol/LNS-398处理hela细胞48h,72h后,细胞增殖明显受到抑制,呈时间和剂量依赖性特点。200umol/LNS-398处理hela细胞48h后,流式细胞仪细胞周期分析表明,NS-398处理组G1期细胞明显增加,S期细胞明显减少。而细胞凋亡指数无明显变化。结论体外实验表明,NS-398能有效抑制宫颈癌细胞生长,其机制可能与其阻滞细胞周期有关。[Objective] To evaluate the anti-prollferative and apoptotic effect of NS-398,a selective COX-2 inhibitor, on human cervical carcinoma cell line Hela in vitro. [Method] Hela ceils were treated with different concentrations of NS-398 for 48 and 72 h. the proliferation of Hela cells was studied by MTT assay. The cell cycle distribution and apoptotic index were detected by flow cytometry analysis. [Results] NS-398 results in significant antiproliferafive effects on hela ceils in dose and time-dependent. After treated with 200 umol/L NS-398 for 48 h, NS- 398 increased the proportion of cells in the G1 phrase and decreased the proportion in the S phrase of the cell cycle. however, the apoptotic index showed no difference. [Conclusion] NS-398 could effectively suppress the growth of hela cells in vitro, the mechanism probably relates to its blockage of cell cycle.
