康欣胶囊预处理对急性缺血性肾衰的保护作用研究

摘  要:目的 研究康欣胶囊对大鼠缺血再灌注肾基因谱的影响,从基因水平研究康欣胶囊的作用机理。方法 SD大鼠随机分为不服用康欣胶囊对照组和服用康欣胶囊实验组,康欣胶囊灌胃5周後,建立缺血模型.采用基因芯片检测康欣胶囊对肾相关基因表达情况的影响。结果 肾4,097个相关基因中,康欣胶囊可使370个基因表达下调,390个基因表达上调;其中肾老化基因活性扫描结果为Cy5:893,CyY3:2,184,Ratio:0.411。实验组缺血7d尿素氮为7.20±0.54mmol/l,肌苷为82.70±10.54umol/l,与对照组比较P〈0.05。结论 康欣胶囊对肾相关基因表达具有下调和上调两方面作用,通过下调肾老化基因表达,延缓肾老化,保护肾脏。To evaluate the effects of the Kanxin Capsule with gene-chip on the rat model of renal ischemia and reperfusion. We divided 24 SD rats into two groups at random: The experimental group served with Kanxin capsule and the blank group without Kanxin capsule. The experimental group was fed with Kanxin capsule for 5 weeks. Made the renal failure by the SD rat model of renal ischemia and reperfusion injury and then detected the gene-chip of the right kidney. 370 gene expressed down-regulation and 390 up-regulation in 4, 097 gene. The result of the renal aging gene is Cy5: 893, CyY3: 2, 184, Ratio: 0.411. In the group of the Kanxin capsule, BUN is 7.20±54mmol/1 and Cr is 82.70±10.54umol/l in 7 days of ischemia and reperfusion injury. Compared with the blank group, p 〈 0.05. Kanxin capsule could make gene expression being down-regulation or up-regulation and has the effect on protecting the renal function by regulating the aging gene down.
