用荧光分光光度法测定血清中的苯丙氨酸及其应用

摘　　要：用荧光分光光度法测定健康成人及苯丙酮尿症（ＰＫＵ）患儿血清中游离苯丙氨酸（Ｐｈｅ）的含量，结果：昆明地区健康成人血清中游离Ｐｈｅ浓度为（０．１２８±０．０４）ｍｍｏｌ／Ｌ，ＰＫＵ患儿的浓度明显升高（０．８０～５．０３ｍｍｏｌ／Ｌ）．结果表明：该法有助于对苯丙酮尿症患儿的诊断．此外，文章还从方法学的角度（荧光光谱、线性相关、重复及回收试验）对该法的可信度及影响荧光测定的因素作了分析和探讨．Fluorescence spectrophotometry was used to determine the values of serumfree phenylalanine(Phe)in healthy adults and child patient with phenylketonuria(PKU).The results were that serum free Phe concentration was(0.128±0.04)mol/L in healthyadults and rose obviously in PKU child patients(0.80±5.03 mmol/L)across the region ofKunming.Hence it could be concluded from the above that PKU child victims acquired agreat benefit from the diagnostic application of this method,whose creditability and influen-tial factors were further discussed from the angle of methodology(fluorescence spectrum,lin-ear correlation,repetition and recovery test).
