牛碱性成纤维细胞生长因子在酵母中表达产物的纯化与鉴定

摘　　要：在以往工作基础上，将已经鉴定的牛碱性成纤维细胞生长因子（ｂｂＦＧＦ）基因克隆至酵母分泌型表达载体ｐＶＴ１０２Ｕ／α，并转化宿主菌Ｓ－７８。表达产物经一系列纯化后，得到ＳＯＳ－ＰＡＧＥ银染鉴定纯度＞９０％的最终产物，免疫学和细胞学检测结果表明，重组ｂｂＦＧＦ具有与天然ｂｂＦＧＦ相同的生物活性。The recombinant plasmid pVT102U/α-bbFGF constructed previously was expressed in the yeastSaccharomyces cerevisiae S-78.The expressed product was purified by chromatography on CM-52 celluloseand affinity chromatography on heparin-Sepharose4B. The immunogenic property of the expressed productwas detected using the ELISA method, and its bioactivity(the stimulating effect on 3T3 cells wasmesured. They both showed to possess the same activity with that of the natural one.
