摘  要:目的:探讨慢性可变应激对大鼠强迫游泳行为的影响及其作用机制。方法:首先用强迫游泳实验对Wistar大鼠进行行为评分,选择得分相近的Wistar大鼠12只,随机分为正常对照组及模型组,每组6只,通过不同形式的慢性应激刺激引起大鼠强迫游泳行为改变,用放射免疫方法测定血浆皮质醇含量变化,镜下观察海马CA3区的结构改变。结果:与正常对照组比较,模型组大鼠在强迫游泳实验中的不动时间明显延长,上窜时间缩短,血浆皮质醇含量明显增高(P<0.01),海马CA3细胞数目明显减少(P<0.05)。结论:慢性可变应激能引起大鼠强迫游泳实验中的绝望行为,此变化与皮质醇分泌增加有关,并使海马CA3区细胞受损。Objective To observe the effect of chronic variable stress on the forced swimming test of rats, and investigate its preliminary mechanism. Methods First a mark was given to each Wistar rat by forced swimming test. 12 Wistar rats which had similar scores were selected and divided into control group and model group averagely. The behavior changes of rats were induced in the forced swimming test by chronic stress and the plasma cortisol level was analyzed through radioimmunoassay (RIA) methods. The changes of hippocampus CA3 structure were observed with microscope. Results Compared with control group, chronic variable stress increased immobility time, reduced climbing time evidently and enhanced the weight of adrenal gland and the level of plasma cortisol (P〈 0. 01). The number of hippocampus CA3 cells was decreased (P〈0. 05). Conclusion Chronic variable stress can induce behavioral despair of rats and this effect relates to the excessive secretion of cortisol and the damage of hippocarnpus CA3.
