内皮抑素、阿霉素联合使用对左肝叶切除小鼠原位移植性肝癌抑制作用的实验研究

摘　　要：目的研究内皮抑素、阿霉素联合使用对肝叶切除小鼠原位移植性肝癌的生长抑制作用及其机理。方法将由小鼠H22肝癌细胞培养成的实体瘤接种于同期切除左肝叶的昆明小鼠肝脏,建立小鼠原位移植性肝癌模型40只,随机分成4组;对照组:隔日腹腔注射生理盐水;内皮抑素组:隔日腹腔注射内皮抑素12mg/kg;阿霉素组:隔日腹腔注射阿霉素1mg/kg;内皮抑素联合阿霉素组:联合使用内抑素和阿霉素,方法:剂量同上述2组。用药2周后处死小鼠,检测小鼠肝原位肿瘤的体积、肿瘤组织的微血管密度(MVD)、凋亡指数(AI)及小鼠处理后肝功能的情况。结果内皮抑素联合阿霉素治疗对肿瘤生长的抑制较单用内皮抑素或阿霉素有显著差异(P<0.05),单用内皮抑素和单用阿霉素对肿瘤的抑制作用有明显差异(P<0.05),单用内皮抑素对肝功能的影响同对照组之间无显著差异(P>0.05),而阿霉素组和联合组对肝功能的影响同对照组有显著差异(P<0.05)。结论内皮抑素联合阿霉素对原位小鼠肝癌移植瘤的抑制有协同作用,内皮抑素对肿瘤的抑制作用不仅比阿霉素明显,而且无明显肝毒性,尤其适用于肝功能受损情况下(如肝叶切除)的早期使用。Objective To investigate the antitumor efficacy of combining endostatin with doxoiubicin and explore its intrinsic mechanism. Methods Hepatocellular carcinoma graft cultivated with H22 carcinoma cell was implanted in forty mice after resecting left liver lobe, which were randomized into 4 groups to received every other day for two weeks before sacrifice peritoneal cavity injection of：control group （nomal saline）, Es group （12mg/kg）, Adm group （1mg/kg）, combined group （Es 12mg/kg,Adm 1mg/kg）. Tumor volumn, intratumoral microvessel density （MVD）, apoptotic indes （AI） and liver function were evaluated. Results The antitumoral effects of combined group were siginificantly different from Es group or Adm group （P〈 0.05）. There was difference between Es group and Adm group. The liver function of Es group was similar to control group （P〉 0.05）, Whereas the liver function of Adm group and combined group was remarkable different from control group （P〈0.05）. Conclusion There were synergistic antitumoral effects by combining Es with Adm. The inhibitory effects of Es were not only better than that of Adm but also no toxicity to liver, specially suitable to be used early under liver disfunction.
