摘 要:目的:初步探讨IκBα磷酸化抑制剂BAY 11-7082对人卵巢癌裸鼠移植瘤生长及血管生成的影响。方法:选用人卵巢癌细胞系COC 1建立裸鼠皮下移植瘤模型,腹腔注射BAY 11-7082,测量移植瘤的重量、体积,并用TUNEL法检测移植瘤细胞凋亡情况。同时应用免疫组化染色,检测瘤组织内的NF-κB p 65、血管内皮生长因子(VEGF)和FⅧ蛋白表达,并计数微血管密度(MVD)。结果:BAY 11-7082组移植瘤重量显著低于阴性对照组(P<0.05),抑制率为47.60%;BAY 11-7082组AI值高于对照组(P<0.05);PI值小于对照组(P<0.05),BAY 11-7082组NF-κB p 65、VEGF与MVD均显著低于对照组(P<0.05)。结论:BAY 11-7082能明显抑制人卵巢癌细胞系COC 1裸鼠移植瘤的生长,通过影响VEGF表达对抗化疗药物。Objective: To study the effect of BAY 11 - 7 082 ( IκBα inhibitor), on the growth and angiogenesis of human ovarian cancer xenografts in nude mice. Methods: COC1 ceils were selected and were injected subcutaneously in thymus nude mice to establish xenograft models. The mice were divided into control group and BAY 11 -7 082 group (n = 6 in each group) . The inhibitory rate was calculated according to the weights of xenografts. And apoptosis rate of xenografts cells was &retted by TUNEL assay. The proteins expressions of NF -κB p 65, vascular endothelial growth factor (VEGF) and F Ⅷ were assessed by immunohistology method, and MVD was counted according to F Ⅷ staining. Results: The weight of xenografts was significantly lower in PDTC group than that in control group (P 〈 0. 05) and the inhibitory rate was 47. 60%. AI in PDTC group was higher than that in the control (P 〈 0. 05), whereas PI was higher in PDTC group than in the e/milo (P 〈0. 05) . The expressions of VEGF and MVD count of were significantly decreased in PDTC group than that in control group (P 〈0. 05) . Conclusion: BAY 11 -7 082 can inhibit the growth of human ovarian cancer COC1 xenografts in nude mice, which may be due to the down regulation of VEGF thus inhibit tumor angiogenesis.
