定点时段高分压氧对体外培养的人牙周韧带细胞增殖和碱性磷酸酶活性的影响

摘  要:目的探讨高分压氧(high partial pressure oxygen,HPPO)对体外培养的人牙周韧带细胞(human periodontal ligament cells,hPDLCs)增殖和分化的影响,为高压氧(HBO)辅助治疗促进牙周组织修复再生提供研究基础和理论依据。方法组织块法体外培养hPDLCs,定点分别以不同分压的HPPO对hPDLCs处理不同时间,观察细胞形态;通过四唑盐比色法(MTT)和酶动力学方法,动态观察HPPO对hPDLCs增殖和碱性磷酸酶(ALP)活性的影响。结果在定点时段HPPO处理后hPDLCs增殖明显,形态良好;下列方案HPPO处理能显著促进hPDLCs的增殖,提高其ALP活性(P<0.05或P<0.01):100 kPa-2 h/d、200 kPa-1 h/d、400 kPa-30 min/d和600 kPa-30 min/d。下列方案处理后,作用相反(P<0.05或P<0.01):100 kPa-6 h/d、200 kPa-4 h/d、400 kPa-4 h/d和600 kPa-2 h/d。结论一定量的HPPO可以促进体外培养的hPDLCs的增殖和分化,HBO辅助治疗可能是促进牙周组织愈合和再生的有效方法之一。Objective To investigate the biological effects of high partial pressure oxygen (HPPO) at vested time and intervals on the proliferation and alkaline phosphatase activity of human periodontal ligament cells in vitro, and to.provide experimental evidence for using hyperbaric oxygen (HBO) in promoting the periodontal tissue regeneration. Methods The fifth passage of human periodontal ligament cells ( hPDLCs ) were cultured in a 5% humid CO2 environment at 37 ℃ by using dulbecco minimum essential medium (DMEM) supplemented with 10% fetal bovine serum (FBS) for 24 hours, then the PDLCs were treated by different oxygen pressure from 100 kPa to 600 kPa for different durations every day. 3-( 4,5-dimethyhhiazol-2-yl )-2,5- diphenyltetrazolium bromide dimethylbenzene (MTT) colorimetric assay and alkaline phosphatase (ALP) activity analysis were used at different time points ( the first, the second, the third, the fifth and the seventh day). Results Under HPPO environment,hPDLCs kept their normal shape,and proliferated more rapidly than normal cultured cells. When compared with the control group, treatment by HPPO at the following pressures and durations ( 100 kPa-2 h/d,200 kPa-1 h/d,400 kPa-30 min/d or 600 kPa-30 min/d) significantly stimulated the proliferation and ALP activity(P 〈0.05 or P 〈0.01 ) ;While after being treated at 100 kPa-6 h/d,200 kPa-d h/d,400 kPa4 h/d or 600 kPa-2 h/d, the proliferation and ALP activity were decreased significantly ( P 〈 0.05 or P 〈 0.01 ). Conclusions HPPO could exert both promotive and inhibitory influence on the proliferation and ALP activity of cultured hPDLCs, and HBO treatment may be an effective way to promote periodontal tissue regeneration.
