摘  要:目的研究重组人白细胞介素24(rhIL-24)及真核表达基因重组质粒pcDNA3·0-hIL-24对黑色素瘤B16细胞(B16细胞)及其荷瘤小鼠体内外抑肿瘤作用。方法用rhIL-24蛋白作用于小鼠B16细胞,检测其对B16细胞的生长抑制作用及诱导凋亡作用,并用rhIL-24蛋白及pcD-NA3·0-hIL-24治疗荷瘤小鼠。结果rhIL-24蛋白对体外培养的B16细胞具有显著的抑制生长和诱导凋亡作用,rhIL-24蛋白及pcDNA3·0-hIL-24对C57/BL6小鼠体内形成的恶性黑色素瘤具有明显的抑瘤作用。结论rhIL-24蛋白具有生物活性,他及真核表达重组质粒对B16细胞及其小鼠种植瘤生长有抑制作用,并能诱导肿瘤细胞凋亡及促进其分化。Objective To study the anti-tumor effects of hIL-24 gene and its products on B16 melanoma cells in vitro and in vivo. Methods rhIL-24 protein and pcDNA3. 0-hIL-24 were used to treat B16 cells in vitro and the tumors in C57/BL6 mouse after B16 cells injection. MTT, flow cytometry and FACS analysis were used to examine the results,and pathological methods were used to check the tumors. Results rhIL-24 protein could strongly suppress the growth of B16 cells, induce its apoptosis in vitro, rhIL-24 protein and pcDNA3. 0-hIL-24 could suppress the growth of the B16 cells' tumors. Conclusion The rhIL-24 protein and pcDNA3.0-hIL-24 could perform its biological functions in vitro and in vivo in suppressing the growth of B16 cells and its tumors and induceing its apoptosis and differentiation.
