摘 要:目的:研究丹参注射液对槟榔提取液(aqueousarecanutextract,AANE)所导致的血管内皮细胞(endothe-lialcell,EC)损伤的保护作用,旨在为丹参注射液治疗口腔黏膜下纤维性变(oralsubmucousfibrosis,OSF)提供理论依据。方法:体外培养人脐静脉EC,分为保护组及预防组2组,分别在AANE作用前后将EC与丹参注射液共同培养,MTT法检测EC增殖,放免法检测EC分泌内皮素(1endothelin-1,ET-1)量的变化。结果:①在AANE干预之后将丹参注射液与EC共同培养,与对照组相比,EC增殖情况明显增加(t=-2.805,p<0.05),分泌ET-1的量显著减少(t=2.965,p<0.05)。②在AANE干预之前先将丹参注射液与EC共同培养,发现EC增殖(t=2.701,p>0.05)分泌ET-1的量(t=-1.541,p>0.05)与对照组相比差异无统计学意义。结论:丹参注射液能有效抑制AANE导致的EC损伤,使EC分泌ET-1减少,其机制可能通过ET-1这个靶标来发挥抗纤维化作用。Objective:To discuss the mechanism of Salvia Miltiorrhizae Injection (SMI)effects on Oral Submucous Fibrosis (OSF) whether through improving the function of the vascular endothelial cell (EC) damaged by aqueous areca nut extract (AANE).Method: EC (from ECV-304) were cultured in vitro and divided into two groups: the protective group and the preventive group. EC were incubated together with SMI before or after EC were influenced by AANE, and we observed the changes of the proliferation of EC and the level of ET-1 secreted by EC.Result: 1. The protective group: SMI was incubated together with EC after which were affected by AANE, we found that the number of EC was larger (t=-2.805,P〈 0.05) and the level of ET-1 was lower (t=2.965,P〈0.05)than those of the contrast. 2. The preventive group: SMI were incubated together with EC before which were affected by AANE, we found that the number of EC (t=2.701,P〉0.05) and the level of ET-1 (t=-1.541,P〉0.05) were resemble with those of the contrast.Conclusion: Salvia Miltiorrhizae could protect EC through alleviating the injury caused by AANE and play its antifibrotic action by affecting the cells' secreting cytokines.
