突触素在糖尿病视网膜细胞表达改变的研究

摘  要:目的探讨突触素(synaptophysin,SYN)在糖尿病视网膜病变(diabeticretinopathy,DR)发病中的作用。方法Sprague-Dawley雄性大鼠尾静脉注射链脲佐菌素造模,以血糖测定和尿糖水平测定进行筛选,正常对照组尾静脉注射等量枸橼酸钠缓冲液。成模后继续饲养4周,取出眼球视网膜组织,连续冰冻切片,用免疫组织化学SABC法染色观察各组大鼠视网膜SYN的表达情况。结果糖尿病大鼠成模前,两组动物的体重、血糖和尿糖检测结果间差异无统计学意义(P>0.05)。成模后4周,糖尿病组与正常组大鼠体重、血糖和尿糖分别为(189.13±18.41)g,(23.72±3.25)mmol/L,(++至+++)和(260.52±32.15)g,(4.95±0.52)mmol/L,(-),两组差异有统计学意义(P<0.01)。SYN在正常组大鼠视网膜节细胞层及内核层均有表达,正常组大鼠视网膜SYN的光密度值为96.31±15.34。糖尿病大鼠视网膜内SYN表达显著减弱,糖尿病大鼠视网膜SYN的光密度值为65.73±8.92。两组相比,差异有统计学意义(P<0.01)。结论SYN随糖尿病大鼠病程延长、病情加重,表达量减少。其表达量的变化很可能是DR发生、发展的重要因素。Objective To explore the possible role of Synaptophysin (SYN) in the pathophysiology of diabetic retinopathy. Methods Streptozotocin (STZ) - induced diabetic rats and normal rats were employed to study the alteration of the SYN in the retina of diabetics by immunohistocbemistry. The weight, blood sugar and urine sugar were also measured before and after model induction in both groups. Results The data of weight, blood sugar and urine sugar indicated no significant difference between the two groups before animal model induction. Four weeks after the induction of diabetes, the differences between the control and the diabetic group in weight, blood sugar and urine sugar were distinct ( P 〈 0.0l ). When compared with the normal retina, a signiilcant decrease of SYN expression was observed in the diabetic rats' retina ( P 〈 0. 01 ). Conclusions Experimental evidence showed down - regulation of SYN expression in retina of streptozotocin - induced diabetic rats under hyperglycemia condition, and it might play a role in the pathophysiology of diabetic retinopathy.
