摘  要:目的观察重组人内皮抑素的抗肿瘤活性及对血管内皮细胞增殖的抑制作用。方法以重组人内皮抑素作用于小鼠移植性肿瘤模型及人癌裸鼠移植性肿瘤,观察其对肿瘤生长的抑制效果,并与传统细胞毒性药物进行比较;同时,以MTT法观察其对血管内皮细胞ECV细胞和结缔组织类成纤维细胞L929细胞的生长抑制作用。结果重组人内皮抑素对小鼠移植性肿瘤Lewis肺癌及人癌裸鼠移植性肿瘤QGY肝癌、A549肺癌、Bcap37乳腺癌均有明显的肿瘤抑制作用;MTT实验显示重组人内皮抑素对血管内皮细胞及结缔组织类成纤维细胞具有生长抑制作用。结论重组人内皮抑素具有一定的抗肿瘤效果,其作用机理可能在于抑制肿瘤血管内皮细胞的增殖。Objective To analyze the inhibitory effect of recombinant human Endostatin on tumor growth and the proliferation of endothelia cells. Methods Xenotransplanted nude mice model with human cancer and experimental implanted tumor mice model were used to evaluate the antitumor activity of rh-Endostatin. The tyro-toxicity drugs were used at the same time to compare with rh-Endostatin. MTT assay was applied to analyze the anti-proliferation activity of rh-Endostatin on human embryo umbilical cord vascular endothelial cell ECV and connective tissue fibroblast cell L929. Result rh-Endostatin was shown to have obvious anti-tumor effect on Xenotransplanted nude mice model with human hepatic cancer QGY, lung cancer A549 and breast cancer Bcap37 and on experimental implanted tumor mice model: Lewis lung cancer. MTT assay showed the inhibition of rh-Enclostatin on the proliferation of ECV and L929. Conclusion rh-Endostatin was shown to have anti-tumor function on Xenotransplanted nude mice model with human cancer and implanted tumor mice model, and the mechanism may lie in the inhibition of forming new blood vessel in tumors.
