摘  要:目的建立小鼠脑卒中后抑郁(PSD)模型并研究黄蜀葵总黄酮(TFA)对其的影响。方法将105只小鼠适应性饲养1周后随机分为假手术组、单纯卒中组、模型组、氟西汀组及TFA低、中、高剂量组,每组15只。模型组、氟西汀组及TFA低、中、高剂量组右侧大脑中动脉阻断(MCAO)30min后再灌注致脑卒中,予以孤养、慢性不可预见的温和性应激(CUMS)2周,制备PSD模型,MCAO术前2d和术后21d分别分别腹腔注射蒸馏水(等量)、氟西汀2.5mg/(kg·d)及TFA40、80、160mg/(kg·d);假手术组每笼5只饲养,单纯卒中组MCAO30min后再灌注致卒中模型后每笼5只饲养,两组蒸馏水腹腔注射方法和剂量同上。观察各组神经症状评分、体重、蔗糖水偏好百分比和血液流变学变化。结果模型组神经功能恢复较慢、体重增加缓慢、蔗糖水偏好百分比显著降低、全血黏度显著增加;与模型组比较,TFA三个剂量组神经功能恢复、体重增加快,蔗糖水偏好百分比高,全血黏度降低。结论MCAO30min后再灌注+孤养+CUMS的方法可造成小鼠PSD模型;TFA可能通过改善PSD小鼠血液流变学促进其神经功能恢复和抑郁行为改善。[Objective] To set up a mouse model of post-stroke depression(PSD)and study the effect of total flavone in abelmosehus manihot (TFA)on it. [Methods] 105 mice were fed adaptively for 1 week,then diVided /andomiy into fake operation,stoke,model, fluoxetlne, high dose TFA, moderate dose TFA and 10w dose TFA group. 15 mice in each group. Set up the mice model of PSD by using Of separated living and chronie unpredictable mild stress (CUMS) on the base of isehemia model of middle cerebral artery occlusion (MCAO) for 30 min. The mice were performed intraperitoneal injection with distilled water, fluoxetine 2.5 mg/(kg, d); and TFA 40, 80,160 mg/(kg· d)respeetively 2 days before MCAO and 21 days after MCAO. Mice in fake operation group were fed with 5 mice in one cage. Mice in stroke group were performed intraperitoneal injeetion with distilled water. Observed the neurological scores, body mass, preference of 1% sucrose and the hemorheology changes. [Results]Neurological function recovery was significantly slow,body mass increased slowly, the preference of 1% sucrose markedly decreased and the whole blood viscosity markedly increased in PSD mice. TFA 160,80, 40 mg/kg could promote neurological function recovery, increase body mass,increase the preference of 1% sucrose and decrease the whole blood viscosity. [Conclusion] Reperfusion 30 min after MCAO+separated living+CUMS can establish the PSD model. TFA can promote neurological function recovery and improve the depressive behaviors of PSD mice by improving abnormal hemorheology.
