摘  要:目的通过观察番茄红素对长波紫外线(ultraviloet A,UVA)反复照射大鼠皮肤中丙二醛(malonic dialdehyde,MDA)形成和基质金属蛋白酶(matrix metalloproteinases,MMPs)mRNA表达的影响,为寻找新的预防光老化方法提供实验依据。方法Wistar大鼠随机分为对照组(C)、UVA照射组(UV)、UVA照射+番茄红素灌胃组(UV+L)。UV组和UV+L组给予UVA照射(每次剂量20J/cm2,每周3次,共12周,累积剂量720J/cm2)。UV+L组予番茄红素油树酯灌胃(每日1次,每次5mg,共16周),C组和UV组相应不给予番茄红素油树酯灌胃。经处理后以半定量逆转录酶-聚合酶链式反应(RT-PCR)法和改良微量硫代巴比妥酸(thiobarbituric acid,TBA)比色法检测各组皮肤中MMP-1、MMP-2、MMP-9mRNA表达水平及MDA含量。结果UVA反复照射后,UV+L组皮肤中MDA含量及MMP-1、MMP-2mRNA表达水平均明显低于UV组(P<0.01;P<0.01),两组间MMP-9mRNA表达水平差异无统计学意义(P>0.05)。结论番茄红素可明显抑制UVA反复照射大鼠皮肤中MMPs mRNA表达和MDA形成,提示系统性应用番茄红素对UVA致大鼠皮肤光老化有拮抗作用。Objective To explore effects of lycopene on ultraviolet A (UVA)-induced malonic dialdehyde (MDA) formation and matrix metalloproteinases (MMPs)mRNA expression in rat skin. Methods Thirty Wistar rats were randomized into three groups comprising control group (C), ultraviolet exposed group (UV), and ultraviolet exposed and lycopene-treated group (UV+L). Rats of UV group and UV+L group were irradiated with UVA radiation. A dose of 20 J/cm2 was given three times weekly for 12 weeks, yielding a total dose of 720 J/cm2. In rats of UV+L group lycopene oil-resin was gavaged before irradiation. A dose of 5rag was given every day for 16 weeks. Lycopene-free oil-resin was gavaged accordingly in rats of C and UV groups. After UVA radiation, MMPs mRNA expression levels and MDA concentration were observed by semi-quantitative RT-PCR and modified thiobarbituric acid (TBA) microcolorimetric methods. Results MMP-1, MMP-2 mRNA expression levels and concentration of MDA in the UV+L group were significantly lower than those in group UV (P〈0.01 ; P〈0.01). MMP-9 mRNA expression levels in the two groups had no significant difference (P〉0.05). Conclusions Lycopene can inhibit UVA-induced skin MMPs mRNA expression and MDA formation, which indicates that systemic use of lycopene may prevent UVA-induced photoaging in rats.
