摘 要:目的用电刺激技术建立大鼠精神分裂症动物模型,测定伏核和额前皮质脑区内多巴胺及其代谢产物含量的变化。方法电刺激大鼠中脑腹侧被盖区,建立精神分裂症动物模型,将大鼠分为正常对照组、假手术组、模型组、氟哌啶醇组,用高效液相色谱-电化学检测法测定不同组别大鼠伏核和额前皮质脑区内多巴胺及其代谢产物含量的变化。结果电刺激大鼠中脑腹侧被盖区引发大鼠类似精神分裂症的阳性和阴性症状;模型鼠伏核内多巴胺含量[(7.56±2.82)ng/mg脑组织湿重]高于正常对照组[(2.10±0.30)ng/mg脑组织湿重](P<0.01),额前皮质脑区内多巴胺含量[(9.81±3.01)ng/mg脑组织湿重]高于正常对照组[(2.97±0.41)ng/mg脑组织湿重](P<0.01);氟哌啶醇组大鼠伏核内多巴胺含量[(2.11±0.10)ng/mg脑组织湿重]低于模型组(P<0.01),额前皮质脑区内多巴胺含量[(3.12±0.43)ng/mg脑组织湿重]也低于模型组(P<0.01)。结论电刺激大鼠中脑腹侧被盖区可建立精神分裂症动物模型,模型大鼠伏核等脑区内多巴胺功能亢进,氟哌啶醇能有效的降低多巴胺含量。Objective To establish animal model of schizaphrenia by electrically stimulating ventral tegmental area (VTA) in rats;and to measure contents of dopamiue ( DA ) and its metaholites in the nucleus aceumhens (NAc) ;and prefrontal cortex (PFC). Methods Animal model of schizophrenia was established successfully hy electrical stimulation . The rats were divided into four group: control group, sham-operated group, model group and HAL group. The contents of DA and ils metabolites in NAe and PFC of schizophrenia rats were detected hy HPLC-EC. Results Abnormal behaviors were induced by high-frequency stimulation in VTA. Contents of DA(7. 56 ± 2.82ng/mg wet weight of brain tissue)in right NAe in model group were higher than those in control group (2.10 ±0.30 ng/mg wet weight of brain tissue) ( P〈 0. 01 ) and contents of DA(9.81 ± 3.01 ng/mg wet weight of brain tissue ) in PFC hi model group were higher than those in control group(2.97 ±0.41 ng/mg wet weight of brain tissue) ( P〈0.01 ). Contents of DA(2. 11 ±0. 10 ng/mg wet weight of brain tissue) in right NAc in HAL group were lower than those in model group ( P 〈 0.01 ). Contents of DA ( 3. 12 ± 0. 43 ng/mg wet weight of brain tissue ) in PFC in HAL group were lower than those in model group( P〈0. 01 ). Conclusion Schizophrenia animal model was established successfully. DA is hyperaetive in NAc and PFC of schizophrenia rats, hut haloperidol (HAL)can effectively reduce DA contents.
