犬瘟热病毒疫苗株附着蛋白基因的克隆及同源性比较

摘　　要：根据发表的犬瘟热病毒(CDV)参考株Ondetstepoort的序列设计1对引物,以犬瘟热病毒疫苗株感染Vero细胞总RNA为模板,利用RT-PCR扩增出附着蛋白基因843 bp片段,将这个片段连接到pMD18-T载体上,经过PCR鉴定、酶切鉴定得到1个阳性克隆,将阳性质粒进行序列测定,结果表明,该片段与Onderstepoort株核苷酸同源性为95.9%。从基因角度为犬瘟热的预防、诊断和治疗提供理论依据。One pair of primers was designed and synthesized based on the haemagglutinin （H） protein gene sequence of the Onderstepoort strain of canine distemper virus（CDV） The total RNA was isolated from the Vero cells line infected with the vaccine strain of CDV. The products of reverse transcription （RT） reaction were used as PCR templates. The H gene fragment was amplified by polymerase chain reaction（PCR）. A 843 bp nucleotide fragment of the PCR products was connected with the vector pMD18 - T. The recombinant plasmid was identified by restriction endonuclease Kpn Ⅰ/Apa Ⅰ and PCR. and then sequenced. The results showed that the H gene sequence of the vaccine strain shared homology of 95.9 %. with that of the Onderstepoort CDV strain
