摘 要:目的探讨端粒酶逆转录酶(hTERT)在年龄相关性白内障患者晶状体上皮细胞(LEC)中的表达及在白内障形成中的作用。方法应用免疫组织化学及核酸原位杂交技术检测hTERT在100例年龄相关性白内障患者和30例正常LEC中的表达,并进行比较。结果年龄相关性白内障LEC中hTERT的阳性表达率明显增高(P<0.05),染色强度以中等以上强度为主,与正常晶状体组相比,差异具有显著统计学意义(P<0.01)。三种不同类型的年龄相关性白内障LEC中hTERT的阳性表达率及染色强度均无显著统计学差异(P>0.05)。结论年龄相关性白内障LEC中端粒酶的激活可能是对紫外线辐射及氧化应激诱发的DNA损伤、端粒缩短的一种防御反应。但当损伤积累到一定程度时,端粒酶的激活也不足以阻止端粒缩短,LEC即发生衰老或凋亡,形成白内障。Objective Telomerase is closely related to cellular immortalization and cancer development,and the research about it in ophthalmic field just reaches a basic stage. This paper was to investigate the expression of human telomerase reverse transeriptase (hTERT)in the lens epithelial cells(LEC) of age-related cataract and its effect on age-related cataract formation. Methods In situ hybridization and immunohistoehemistry techniques were used to detect the mRNA and protein expression of hTERT in 100 LEC specimen of age-related cataract,20 LEC specimen from traumatic cataracts and other 10 transparent lens of high myopia. Results The positive rate of hTERT in age-related cataract LEC(79% ) was significantly higher with the moderate and strong staining intensity in comparison with traumatic LEC specimen and clear LEC specimen( positive rate,40% ) with significant differences among them ( P 〈 0. 01 ) , showing brown-yellow or brown staining granular. There were no statistically significant differences in the positive rate and staining intensity of hTERT among the cortical cataract,nuclear cataract and posterior subeapsular cataract (P 〉 0. 05). Conclusion Overexpression of hTERT in age-related cataract LEC is a response to DNA damage. That increase of telomerase activity is not enough to prevent decrease of telomere length is the main mechanism of age-related cataract.
