摘  要:目的:探讨谷胱甘肽乙酯滴眼液抑制糖尿病性白内障的效果及作用机制。方法:复制链脲佐菌素(streptozocin,STZ)糖尿病大鼠模型,成模前药物治疗组每日给予2次谷胱甘肽乙酯滴眼液治疗,而对照组给予磷酸钠缓冲液。采用英国牛津大学晶状体分级方法记录晶状体混浊的程度。并于实验过程中观察大鼠的一般状况,了解还原型谷胱甘肽乙酯对糖尿病大鼠一般状况的影响。结果:糖尿病诱导的晶状体混浊进展呈双向趋势:前6wk缓慢增长,后7wk快速进展。治疗组大鼠的混浊进展相对于未治疗组较慢,并且在第4wk时两者差别具有统计学意义(治疗组vs未治疗组,P<0.05)。同时药物治疗组大鼠与未治疗组相比较,精神、毛色等一般状况较好。结论:谷胱甘肽乙酯滴眼液抑制了早期晶状体混浊的进展,它作为合成谷胱甘肽的前体,进而发挥其抗白内障的作用。AIM: To evaluate the effect of glutathione ethyl ester (GSH-EE) drops on the progression in diabetic cataract. METHODS: Diabetic models were made by treatment of rats with streptozotocin (STZ). Rats were divided into three groups: normal group, drug-treated group and untreated group. The drug-treated rats were pre-dropped with 0.1% GSH-EE drops before they were injected with STZ, and those in the untreated rats received sodium phosphate buffer solution, twice daily. Transparency of lenses was observed weekly by using a slit lamp, as well as the rats' general conditions in order to evaluate the effect of GSH-EE. " RESULTS: The progression in lens opacity induced by diabetes showed a biphasic pattern. An initial slow increase in the first 6 weeks followed by a rapid increase in the next 7 weeks. The progression in lens opacity in the treated group was slower than that in the untreated group, with significant difference between the treated and untreated groups (P〈0.05) at the 4th week. The rats' general conditions, such as general condition and skin color, were better in the treated group than that in untreated one. " CONCLUSION: GSH-EE can inhibit the progression of the diabetic cataract at the earlier stage. It may maintain the lens transparency and function by serving as a precursor for glu-tathione biosynthesis and protecting the sulfhydryl groups from oxidation.
