摘  要:目的建立小鼠肝脏肉芽肿模型及探讨其机制。方法痤疮丙酸杆菌(P.acnes)经尾静脉注射给C57BL/6J(B6)小鼠。在注射前、注射后1、6 h及1、2、3、5、7 d分别检测外周血F4/80^-B220^- CD11c^+细胞含量及ALT(丙氨酸转氨酶)水平并作肝脏组织病理学检查。进而将外周血分离出来的F4/80^-B220^-CD11c^+细胞经尾静脉注射给同种正常小鼠,建立过继转移模型,观察其ALT水平及肝脏组织病理学变化。结果注射P.acnes的小鼠外周血F4/80^-B220^-cD11c^+细胞含量及ALT水平明显升高,病理学观察显示肝脏有大量炎症细胞浸润并形成肉芽肿。F4/80^-B220^-CD11c^+细胞过继转移的小鼠ALT水平亦明显升高,肝脏病理切片发现有肉芽肿形成,与注射P.acnes的小鼠肝脏病理切片结果相一致。结论P.acnes能成功诱导建立小鼠肝脏肉芽肿模型,且P.acnes注射后升高的F4/80^-B220^-CD11c^+细胞是诱导形成肝肉芽肿的主要因素。Objective To establish a mouse model of liver granuloma and investigate its mechanism. Methods C57BL/6J mice were injected Propionibactenium aches (P.acnes) via tail vein. After injection, number of F4/80-B220-CD11c^+ cells and level of ALT were detected at time points of 1, 6 h, and 1, 2, 3, 5 and 7 d. On the 7th day after injection, pathological changes of livers between treated mice and untreated normal mice were compared. Nad've mice were injected with the sorted F4/80-B220-CD11c^+ cells from peripheral blood of P.acnes-treated mice to establish an adoptive transfer group, Results Number of F4/80-B220-CD11c^+ cells and level of ALT upgrade obviously after F4/80-B220-CD11c^+ cell infusion as well as P.acnes injection. Pathologic examination showed that granulomas were formed in the livers either in P.acnes or in F4/80-B220-CD11c^+ cell induced mice, accompanied with inflammatory cells infiltration. Conclusion P. acnes can induce liver granuloma of mice successfully and F4/80-B220- CD11c^+ cell play an important role in liver granuloma development.
