摘  要:目的 研究呼吸道合胞病毒(RSV)感染豚鼠后肺组织β肾上腺素能受体(β受体)和M胆碱能受体(M受体)的变化。方法 应用放射性配基结合分析法,测定病毒接种组和对照组豚鼠肺组织β、M受体密度和亲和力的变化。结果 ①RSV接种后豚鼠肺组织均出现毛细支气管炎、肺炎的病理改变。②β受体最大结合容量(Bmax)在RSV接种后无明显改变。β受体平衡解离常数(Kd)在RSV接种后3d增高,5~7d达高峰,以后逐渐下降,接种后14、21 d与对照组比较差异无统计学意义。③高亲和力M受体亚型的Bmax和Kd值在感染前后无明显变化。低亲和力M受体亚型在RSV接种后3d出现Bmax增高,Kd值降低,5—7d达高峰,第21天与对照组差异有统计学意义。结论RSV感染可使β受体亲和力下降,低亲和力M受体密度增加、亲和力上升,两种受体系统之间的平衡失调是引起气道高反应性的机制之一。Objective To investigate the effects of respiratory syncytial vires (RSV) infection on β-M- receptor density and function in lung tissue of guinea-pig. Methods After intranasal inoculation of guinea-pig with RSV, the vires rapidly proliferated in lung tissue. By using the method of specific binding of isotopic ligand to β-M- receptor,the density and affinity of β-M-receptor in lung tissue were measured on days 3,5,7,9,11,14 and 21 after inoculation. Results ( 1 ) The RSV-inoculated group showed pathological evidence of acute bronchiolitis on the 3^rd day after inoculation,which reached the peak during the days 5 to 7 and recovered on day 14. (2) Viresinfection had no significant effect on the maximum binding capacity(Bmax) of β-adrenoceptor in lung tissue, and the dissociation constant of β-adrenoceptor was increased on day 3 postinoculation with RSV, peaked on day 5 to 7, which subsided by day 14,21. (3) Vires-infection had no significant effect on the B max and the Kd of the highavidity subgroup of M-cholinergic receptor. But the B max and the Kd of the low-avidity subgroup of M-cholinergic receptor appeared to increase and decrease on day 3 respectively, which reached the peak on day 5 to 7 and still showed high difference on day 21 .Conclusion The RSV infection could change the density and the affinity of β- M-receptor,which may play a role in airway hyperreactivity.
