摘  要:目的探讨丝裂霉素C(mitomycinC,MMC)与C2-神经酰胺(C2-ceramide,C2-cer)联合应用对人膀胱癌T24细胞生长的抑制作用。方法采用四甲基偶氮唑蓝[3-(4,5-dimethylthiazole-2yl)-2,5-diphenylte-trazoliumbromid,MTT]比色法测定MMC和C2-cer单独或联合应用对人膀胱癌T24细胞生长的抑制作用,并应用Chou-Tala-lay联合指数法定量分析药物之间相互作用的效果。结果MMC、C2-cer单独应用时,随药物浓度增加对T24细胞生长的抑制作用也增加,中效浓度分别为153.426、27.797μmol/L。MMC与C2-cer联合应用时,相互作用的效果表现为在低浓度(0<fa<43%)时呈协同作用(CI<1),高浓度(43%<fa<100%)时呈拮抗作用(CI>1),中效浓度为89.536μmol/L(其中MMC74.048μmol/L,C2-cer15.488μmol/L)。结论MMC和C2-cer相互作用的效果与药物浓度相关,低浓度时呈协同作用,高浓度时呈拮抗作用。OBJECTIVE: To explore the anti-cancer effects.of mitomycin C (MMC) and C2-ceramide (C2-cer) on human bladder cell line T24. METHODS: The MTT method was used to detect the inhibition rate and the combination index method of Chou and Talalay was used to quantitatively assay the effect of combined application. RESULTS: The anti-cancer effects of MMC and C2-cer were enhanced with the increased drug concentrations when they were used alone. The respective median-effect concentrations were 153. 426 and 27. 797 μmol/L. MMC was synergetic with C2-Cer (CI〈1) at low concentrations (0%〈fa 〈43%) and was antagonist with C2-Cer (CI〉1) at high concentrations (43 % 〈 fa〈 100 % ). The median-effect concentration was 89. 536 μmol/L (MMC 74. 048 μmol/L, C2-cer 15. 488 μmol/L). CONCLUSIONS: The effects of combination between MMC and C2-cer are relevant to their concentrations. MMC is synergistic with C2-cer at lower concentrations and antagonistic at higher concentrations.
