摘  要:目的探讨苯乙酸钠(NaPA)对小鼠体内种植性肿瘤的作用及对c-myc基因和免疫力的影响,寻找新的有效的肿瘤治疗药物。方法制作小鼠体内种植性肿瘤模型,NaPA腹腔内注射10 d后运用反转录-多聚酶链式反应(RT-PCR)方法检测肿瘤细胞的c-myc基因扩增情况,应用流式细胞仪(FCM)检测小鼠的免疫功能,同时检测NaPA对荷瘤小鼠的抑瘤作用和毒副作用。结果随着NaPA浓度的增加,对荷瘤小鼠的抑瘤作用加强,且明显抑制c-myc基因的表达,并具有提高小鼠免疫力的趋势。没有检测到对荷瘤小鼠正常组织的毒副作用。结论NaPA可抑制荷瘤小鼠c-myc基因过度表达,抑制肿瘤细胞恶性增殖,并且无毒副作用,有望成为新的肿瘤治疗药物。Objective In order to find a effective anti-tumor drig,we investigate NaPA' s influence on c-myc gene and immunity function in mouse in which tumor was planted. Methods After making a model of mouse in which carcinoma was planted,NaPA was injected in abdominal cavify of those mouse. After 10 days ,we detected the amplified c-myc gene with RT -PCR and immunity function with FCM. At the same time,NaPA' s inhibition of tumor and side effects on the mouse were checked. Results With the density of NaPA added ,the inhibition on tumor can be strengthened and the mouse immunity function can be increased. C-myc gene expression was restrained. We didn' t find side effect on normal organs. Conclusions NaPA can effectively testrain the c-myc gene overexpression and carcinoma reproducing. Na.PA has little side effect,it can be a new anti-tumor drug in the future.
