安更胶囊治疗女性更年期综合征的药效学研究

摘　　要：目的观察中药安更胶囊用于女性更年期综合征的实验治疗效果。方法采用戊巴比妥钠诱导小鼠睡眠实验,观察安更胶囊的协同促睡眠作用;采用醋酸所致小鼠扭体法,观察安更胶囊的镇痛作用;采用毛果芸香碱诱导小鼠发汗实验,观察安更胶囊的抑制发汗作用;采用小鼠强迫游泳实验和悬尾实验,观察安更胶囊的抗抑郁作用;采用雌性大鼠去卵巢模型,观察安更胶囊对大鼠血清雌激素、雄激素、LH和FSH激素水平的影响。结果安更胶囊剂量大于0.41 g.kg-1(生药量)能显著延长戊巴比妥钠诱导小鼠的睡眠时间;剂量大于1.65 g.kg-1(生药量)能够显著减少冰醋酸所致的小鼠扭体次数并明显减轻毛果芸香碱所致的小鼠发汗作用;剂量大于3.3 g.kg-1(生药量)能够显著减少小鼠强迫游泳不动时间和小鼠悬尾不动时间;安更胶囊对去卵巢大鼠外周血清中E2,T和FSH含量无显著影响,使LH含量显著降低,对去卵巢大鼠子宫指数无显著影响。结论安更胶囊有促小鼠睡眠、抗抑郁和较强的镇痛作用;对支配汗腺的交感神经兴奋具有一定的抑制作用,并无雌激素样作用。OBJECTIVE To evaluate the pre-clinical effect of Angeng capsules on treatment of menopausal syndrome in mice and rat models. METHODS The sleep enhancement action with pentobarbital sodium （PS） was observed in pre-treated mice model.The analgesic effect was observed with writhing test. Antidiaphoresis action was observed with pilocarpine in pre-treated mice model. The antidepressant action was evaluated with forced swimming test and tail suspension test.The effect on the serum level of estradiol, androgen, LH and FSH was determined in castrated female rat. RESULTS Angeng capsules（ 〉 0.41 g·kg^-1 ） significantly enhanced the sleeping time in mice pre-treated with PS. When the dose was above 1.65 g·kg^-1, Angeng capsules significantly reduced the frequency of writhing and diaphoresis; Angeng capsules （〉 3.3 g·kg^-1） significantly increased swimming time in forced swimming test and moving time in tail suspension test. Angeng capsules showed no significant effect on the E2, T and FSH level in rat serum, but made a significant decline of LH level in the serum. CONCLUSION Angeng capsules can promote sleep, exert antidepressant, antifatigue and analgesic effect, and inhibit the secretion of sweat glands without decreasing serum estrodial level. It＇s a promising anti-menopausal syndrome Chinese herbal medicine.
