摘 要:以陈放一年的油菜花粉为原料,比较了油菜花粉的两种破壁方法99温差破壁法和蒜汁酶破壁法,通过比较破壁前后各组分蛋白含量以及抗氧化活性确定了球蛋白为目的蛋白,并在此基础上确定了球蛋白的最佳提取条件。研究结果表明,目前条件下温差破壁法可达到实验目的,破壁脱脂后的油菜花粉中主要是球蛋白和谷蛋白;球蛋白的最佳提取条件是氯化钠浓度0.20mol/L,pH值为5,浸提时间6h,浸提次数3次,浸提温度保持4℃。Rape pollen which was stored more than 1 year was used as material in this study. The effect of temperature difference cell-fragmentated method was compared with hydrolization with enzymes from juice of garlic. A research on protein fractionation of rape pollen defatted and cell-fragmentated and their antioxidant effect were performed. On the basis of study,the optimum conditions of extraction of globulins was gained. The results showed that the temperature difference was best choice,glutelins and globubins were found to be the predominant proteins in rape pollen, Through the orthogonal test,the optimum conditions of extraction have been obtained as follows: nacl concentration 0.20mol/L, pH value 5, time 6h, times 3, temperature 4℃.
