摘  要:目的探讨内源性转化生长因子β1(TGF-β1)在心肌缺氧缺血—再灌注损伤修复及心肌重塑中的作用。方法根据新生鼠常压窒息模型,制作缺氧缺血—再灌注损伤大鼠动物模型,将60只新生大鼠随机分成缺氧缺血—再灌注损伤1d组(A组)、7d组(B组)、14d组(C组)以及正常对照组(D组)4组。每组15只用免疫组化方法对TGF-β1在心肌的分布及表达进行定性及半定量分析,用Masson染色方法测定心肌胶原容积(CVF),同时利用HE染色观察心肌组织的病理学改变。统计分析应用SPSS11·0统计软件进行。多组均数比较采用单因素方差分析,两变量之间的关系采用相关分析。结果TGF-β1表达:B组及C组分别与对照组相比,均有统计学意义(P均<0·01),胶原含量(CVF):B组及C组较正常对照组差异有统计学意义(P均<0·01),相关分析表明,CVF与TGF-β1之间呈正相关(r=0·574,P<0·01)。结论大鼠缺氧缺血—再灌注损伤后TGF-β1表达量随损伤时间不同而变化,其可能参与缺氧缺血—再灌注所致的内脏损伤后的自我修复。缺氧缺血—再灌注损伤后TGF-β1的表达与心肌胶原含量的变化呈正相关,提示TGF-β1可能参与损伤后心肌重塑。Objective To investigate the role of TGFβ1 in cardiac self-repairing and remodeling after injuries due to ischemia-reperfusion. Methods The rat model of ischemia-repeffusion injuries was used. Neonatal rats (7-10 days old) were randomly divided into four groups: 1-,7-, 14-day ischemiareperfusion injury group and normal control group. Expression and distribution of TGFβ1 in cardiomyocytes was examined by qualitative and semiquantitative immunohistochemistry. Masson staining was used in the study of total collagen volume fraction (CVF). All calculation were performed with SPSS11.0. Statistical analysis for comparison of data between groups was performed by the one-way analysis of variance. Correlation analysis was performed using pearson correlation coefficient. Results TGFβ1 gene expression increased markedly at 7-day group (P 〈 0. 01 ), and peaked at 14-day group (P 〈 0. 01 ). Compared with the normal control group, CVF increased significantly in 7-day and 14-day ischemiareperfusion injury groups. Correlation analysis revealed that CVF was positively correlated with TGFβ1 ( r = 0. 574, P 〈 0. 01 ). Conclusions TGFβ1 may play an important role in cardiac self-repairing and remodeling after injuries due to ischemia-reperfusion.
