摘 要:目的:了解我国青春前期女童骨钙素基因多态性分布并探讨其对骨量的影响。方法:选择152名9~11.5岁未月经初潮的健康女童,采用双能X线骨密度仪(DXEA)检测对象全身、L1-L4腰椎、左侧近端股骨的骨矿含量、骨密度以及体成分,采用PCR-RFLP技术检测骨钙素基因多态性。结果:152名女童OC基因HindⅢ位点多态性基因型的分布频率分别为:突变型纯合子(HH)5.26%,杂合子(Hh)36.84%,野生型纯合子(hh)为58.90%,各基因型TBBMC、全身及各部位的BMD、血骨钙素水平均存在显著差异(P<0.05),表现为HH<Hh<hh的趋势。结论:我国正常青春前期女童OC基因多态性分布与骨量有关,HH基因型TBBMC、TBBMD、FNBMD显著低于Hh和hh基因型。Objective: To assess the proportion of genotypes of osteocalcin gene polymorphism, and to investigate if the polymorphism of osteocalcin gene is associated with BMD in Chinese premenarche girls. Methods: 152 healthy premenarche girls of Han ethic (9-11.5y) were recruited. BMD and BMC at the lumber spine (L1-L4), proximal femur, and total body were measured by DXA; PCR-RFLP was used to amplify the polymorphisms of the osteocalcin gene. Results: The proportion of genotype of OC gene polymorphism were 5.26% HH, 36.84% Hh, and 58.90% hh, respectively. Significant differences were observed in BMD and BMC at the total body, and BMD at the L1-L4 lumber spine and femur neck among subjects with different genotypes (P〈0.01). Subjects belonging to HH genotype had significantly lower ostocalcin, BMD and BMC in 11 sites than those with genotypes of Hh and hh (P〈0.01). Conclusion: The polymorphism of ostocalcin gene is associated with bone mineral density in Chinese premenarche girls. The H allele of OC gene polymorphism is a potential predisposing factor of low bone mineral density.
