摘  要:【目的】利用cDNA微阵列技术分析p53、BRCA2、EBI2基因在肺鳞癌组织中的表达概况,为肺癌的早期诊断与治疗提供可能的理论依据。【方法】利用6例分期为Ⅰ期的手术切除肺鳞癌标本与6张含4096个基因的人基因表达谱芯片,将来源于肿瘤细胞与正常肺组织的RNA进行配对,用不同的荧光染色剂进行逆转录标记成cDNA探针后混合,与芯片上的基因进行杂交,通过扫描两者的信号强度,获得差异表达基因。找出在此基因表达谱中均上调或下调的基因。【结果】p53在全部6个肺鳞癌标本中均表现上调,而BRCA2及EBI2在全部6个肺鳞癌标本中均表现下调。【结论】p53、BRCA2及EBI2在早期肺鳞癌中出现差异表达,提示其可能为肺鳞癌的分子标志物,为进一步研究肺鳞癌的发生、发展的分子机制提供了线索。[Objective]To screen the differential expression of genes in stage Ⅰ lung squamous cell carcinoma by cDNA microarrays, and mainly study p53. BRCA2 ,EBI2 genes expression as possible molecular markers of lung squamous carcinoma, and provide possible theoretical basis for its early diagnosis and treatment. [Methods]Six surgical specimens and 6 microarray chips, each containing 4096 human target genes, were used for this study. The pathologic stage was Ⅰ(AJCC) in six patients, six tumor specimens were lung squamous cell carcinomas, paired mRNAs from carcinoma cells and normal lung tissue specimens from the same lobe were labeled with different fluorochromes during cDNA probe synthesis in a reverse-transcription reaction. Both synthesized, labeled cDNA probes were mixed and hybridized to the microarray. The signal intensity of each spot was measured by laser scanner and 6 gene expression profiles were obtained, and further to find out up-regulated (over-expressed) and down-regulated (under-expressed) gene in six specimens. [Results] BRCA2.EBI2 were down-regulated and p53 was up-regulated in all 6 lung squamous cell cancer tissues. [Conclusion]BRCA2.EBI2.p53 are the differential expression genes in stage I lung squamous cell carcinoma, suggesting that they are possible molecular markers and these genes provide important clues for further study of carcinogenesis.invasion.metastasis of lung squamous cell carcinoma.
