摘 要:利用陈旧皮张的DNA提取技术从鸡内金、鸭内金中提取DNA,通过特异扩增DNA片段的聚合酶链式反应(PCR),以线粒体DNA细胞色素b(cyt-b)通用引物中的L14841和H15149为引物扩增DNA片段,将扩增后的DNA 用双脱氧链终止法测定其序列,所测序列分别与文献报道的鸡、鸭序列一致。所得结果证明鸡内金的DNA序列与鸭内金的DNA序列有明显差异,以此能准确区分鸡内金和鸭内金。开创了将分子遗传学技术引入动物药材鉴定的先例,增加了动物药材的鉴定技术,使其鉴定手段提高到分子水平,以增加鉴定结果准确性。并探讨了将此技术引入以动物组织、器官入药的动物药材鉴定中的可行性。A reliable apprisal method to the animal tissue of Chinese traditional medicine was first reported in this paper. The entire DNA of chicken and duck brissons were collected by the technique of Hinguchi's amplified via the polymerase chain reaction(PCR) with the common primers, L14841 and H 15149, and sequenced by the dideoxy method. Both DNA sequences of chicken and duck brissons are the same as Desjardins and Morais (1990) and Korenegay (1993). The results showed that the DNA sequence of chicken brisson is different from that of duck. This study first introduced the molecular genetic technique into the area of apprisal of the animal tissue of Chinses traditional medicine. The apprisal result from the technique of molecular genetic are much more accurate than that of the traditional method.
